用琼脂糖(agarose)将血红蛋白(Hb)固定在热裂解石墨电极表面,制备了Hb-agaose膜修饰电极.包埋在琼脂糖中的血红蛋白与电极直接传递电子.在磷酸盐缓冲溶液中得到一对可逆的血红蛋白辅基血红素Fe(Ⅲ)/Fe(Ⅱ)电对氧化还原峰,式电势为-0.299 V(vs SCE),式电势随缓冲溶液pH值增加而负移且成线性关系,直线斜率为-44 mV/pH,说明血红蛋白的电子传递过程伴随有质子的转移.并研究了Hb-agarose膜修饰电极对H2O2的电催化性质.  相似文献

用海藻酸钠(Sodium Alginate,SA)将肌红蛋白(Mb)固定在热裂解石墨电极表面,制备了Mb.SA膜修饰电极.包埋在SA膜中的Mb在磷酸盐缓冲溶液(PBS)和乙醇混合溶液中与电极直接传递电子,得到一对对称的Mb辅基血红素Fe(Ⅲ)/Fe(Ⅱ)电对的氧化还原峰,式电势为-0.339V(vs SCE).式电势随PBSpH值增加而负移且成线性关系,直线斜率为-47.0mV/pH,说明肌红蛋白的电子传递过程伴随有质子的转移.并研究了Mb-SA膜修饰电极在PBS和乙醇混合溶液中催化还原H2O2和催化六氯乙烷脱氯,该修饰电极可用于H2O2和六氯乙烷的定量检测.  相似文献

介绍了一种简单、新颖的途径,气-液界面组装银纳米粒子成为二维纳米粒子膜.这种纳米粒子膜能有效地固定血红蛋白(Hb)到玻碳电极上.银纳米粒子膜作为电子传递的桥梁,在没有任何媒质的帮助下,能极大促进Hb和修饰的玻碳电极之间的电子传递.在pH值为7.0的磷酸盐缓冲溶液中,Hb显示一对清晰的氧化还原峰,式量电位(E0’)大约为-0.525 V(相对Ag/AgCl,KCl饱和溶液).紫外-可见光谱研究表明,固定后的Hb的生物活性保持不变.固定在银纳米粒子膜修饰的玻碳电极上的Hb可以用作生物传感器检测H2O2.当H2O2浓度从5.0μm升高到1.74 mM时,稳定的电流响应随之线性增加.H2O2的检测限(3σ)是4.3×10-7M.  相似文献

用紫外-可见和红外光谱及原子力显微法表征了血红素蛋白质-甲基纤维素膜修饰电极。紫外-可见和红外光谱显示,在甲基纤维素(MC)凝胶中,蛋白质保持原始构象;在溶液pH3.0—10.0范围内,蛋白质的构象可逆地变化。原子力显微图像表明蛋白质与MC水凝胶之间存在较强的作用。研究了血红素蛋白质催化还原O2、H2O2和NO的机理。实验表明,还原O2和NO时,血红素蛋白质-MC修饰电极能重复使用,催化活性几乎不变;而催化H2O2时,催化活性下降较快,表明其反应历程有显的差异。稳定的蛋白质-MC修饰电极能定量测定H2O2和NO2^-。  相似文献

利用Nafion膜将血红蛋白(Hb)和银纳米粒子(AgNPs)固载到玻碳电极表面制作N O-2生物传感器.循环伏安实验表明:Hb/AgNPs/Nafion膜电极在pH值为6.9的PBS缓冲溶液中呈现出一对明显的HbFe(Ⅲ)/(Ⅱ)氧化还原峰,该电极对N O2-有良好的催化作用,线性范围为2.0×10-5~3.4×10-4 mol/L(n=18,R=0.997),检测限为1.2×1 0-6 mol/L(信噪比为3);故该膜电极可做N O-2生物传感器.  相似文献

研究了具有平面环状共轭体系的硫杂环化合物噻吩羧酸在金表面的自组装单分子膜及其修饰金电极的电化学行为.以Fe(CN)6^4-/3-为“探针”离子,用循环伏安法对自组装膜的形成、电极界面电容及其组装机理进行了研究.结果是噻吩羧酸在Au上的覆盖度约为95%,金电极的界面微分充电电容由修饰前的0.294μF/cm^2下降到0.167μF/cm^2,由此可见噻吩羧酸在金表面形成了致密的单分子膜.  相似文献

用直接滴涂法将血红蛋白(Hb)固定到戊二醛(GA)膜修饰的金电极表面,通过循环伏安法研究了Hb在GA膜修饰金电极上的直接电化学行为.在0.1 mol/L磷酸盐缓冲溶液(PBS,pH 6.00)中,扫描速度为100mV/s时,该修饰电极的峰电位差ΔE=44 mV,氧化还原峰电流之比Ip,a/Ip,c=1.17,说明Hb在GA膜修饰电极上的氧化还原过程是可逆过程;随着缓冲溶液pH值的增大,其峰电位不变,而峰电流呈增大(pH=4.00~6.00)或减小(pH=6.00~8.00)的趋势.通过GA膜对Hb的吸附固定,不仅保持了Hb的生物活性,而且实现了Hb与电极之间的直接电子转移.利用此修饰电极,研究了Hb与药物氨茶碱之间的相互作用,求得Hb和氨茶碱的结合数为2.  相似文献

采用紫外光照射法在咪唑类离子液体[C12min][Br]中一步合成了Au-[C12min][Br]纳米复合物,将该纳米复合物固定牛血红蛋白(Hb)后修饰于玻碳(GC)电极表面构成Hb/Au-[C12min][Br]/GC修饰电极。电化学研究表明,Hb在Au-[C12min][Br]纳米复合物膜上发生了准可逆的电极反应,氧化峰电位(Epa)和还原峰电位(Epc)分别为-0.253V和-0.335V。Au-[C12min][Br]纳米复合物对Hb的直接电子转移有很好的促进作用,表现出良好的电化学活性。Hb/Au-[C12min][Br]/GC对H2O2有较好的电催化能力,对进一步研究H2O2传感器具有重要意义。  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

本文叙述了对海南岛及其毗邻大陆边缘白垩纪到第四纪地层岩石进行古地磁研究的全部工作过程。通过分析岩石中剩余磁矢量的磁偏角及磁倾角的变化,提出海南岛白垩纪以来经历的构造演化模式如下:早期伴随顺时针旋转而向南迁移,后期伴随逆时针转动并向北运移。联系该地区及邻区的地质、地球物理资料,对海南岛上述的构造地体运动提出以下认识:北部湾内早期有一拉张作用,主要是该作用使湾内地壳显著伸长减薄,形成北部湾盆地。从而导致了海南岛的早期构造运动,而海南岛后期的构造运动则主要是受南海海底扩张的影响。海南地体运动规律的阐明对于了解北部湾油气盆地的形成演化有重要的理论和实际意义。  相似文献

The explosive growth of the Internet and database applications has driven database to be more scalable and available, and able to support on-line scaling without interrupting service. To support more client's queries without downtime and degrading the response time, more nodes have to be scaled up while the database is running. This paper presents the overview of scalable and available database that satisfies the above characteristics. And we propose a novel on-line scaling method. Our method improves the existing on-line scaling method for fast response time and higher throughputs. Our proposed method reduces unnecessary network use, i.e. , we decrease the number of data copy by reusing the backup data. Also, our on-line scaling operation can be processed parallel by selecting adequate nodes as new node. Our performance study shows that our method results in significant reduction in data copy time.  相似文献

R-Tree is a good structure for spatial searching. But in this indexing structure,either the sequence of nodes in the same level or sequence of traveling these nodes when queries are made is random. Since the possibility that the object appears in different MBR which have the same parents node is different, if we make the subnode who has the most possibility be traveled first, the time cost will be decreased in most of the cases. In some case, the possibility of a point belong to a rectangle will shows direct proportion with the size of the rectangle. But this conclusion is based on an assumption that the objects are symmetrically distributing in the area and this assumption is not always coming into existence. Now we found a more direct parameter to scale the possibility and made a little change on the structure of R-tree, to increase the possibility of founding the satisfying answer in the front sub trees. The geographic information service is enabled by the advancements in general Web service technology and the focused efforts of the OGC in defining XML-based Web GIS service. Based on these models, this paper addresses the issue of services chaining,the process of combining or pipelining results from several interoperable GIS Web Services to create a customized solution. This paper presents a mediated chaining architecture in which a specific service takes responsibility for performing the process that describes a service chain. We designed the Spatial Information Process Language (SIPL) for dynamic modeling and describing the service chain, also a prototype of the Spatial Information Process Execution Engine (SIPEE) is implemented for executing processes written in SIPL. Discussion of measures to improve the functionality and performance of such system will be included.  相似文献

Advances in wireless technologies and positioning technologies and spread of wireless devices, an interest in LBS (Location Based Service) is arising. To provide location based service, tracking data should have been stored in moving object database management system (called MODBMS) with proper policies and managed efficiently. So the methods which acquire the location information at regular time intervals then, store and manage have been studied. In this paper, we suggest tracking data management techniques using topology that is corresponding to the moving path of moving object. In our techniques, we update the MODBMS when moving object arrived at a street intersection or a curved road which is represented as the node in topology and predict the location at past and future with attribute of topology and linear function. In this technique, location data that are corresponding to the node in topology are stored, thus reduce the number of update and amount of data. Also in case predicting the location,because topology are used as well as existing location information, accuracy for prediction is increased than applying linear function or spline function.  相似文献
