目的:为制备重组小鼠Pem(以下简称mPem)-谷胱甘肽巯基转移酶(GST)融合蛋白,作为研究mPem蛋白功能的材料,方法:根据携带小鼠Pem基因编码序列的模板质粒pEGFP/mPem设计合成特异性引物,PCR扩增小鼠Pem基因编码序列,并插入融合蛋白原核表达载体pGEX-4T-3中,得到重组表达质粒pGEX-4T-3/mPem,用此重组质粒转化大肠杆菌BL21细胞,IPTG诱导重组菌表达mPem蛋白,SDS-PAGE及Western Blot鉴定表达产物。结果:重组菌株明显诱导表达出预期相对分子质量49000的融合蛋白。结论:成功构建了mPem-GST原核表达质粒,并在大肠杆菌中表达出mPem-GST融合蛋白,为mPem蛋白功能的研究打下了基础。  相似文献

为了获得高纯度的可溶性NF-κB相互作用多肽,首先以酵母双杂交技术筛选获得的NF-κB相互作用多肽的酵母表达质粒pGAD GH/pp10为模板,扩增多肽基因片段,后经BglⅡ,StuⅠ双酶切后连接到pET-42a(+)载体中,构建GST/多肽融合蛋白的原核表达载体,并将GST/多肽融合蛋白的原核表达载体转化入BL-21菌株,用0.4 mmol/L IPTG于30 ℃诱导表达4 h,后经GST亲和纯化GST/多肽融合蛋白,SDS-PAGE电泳鉴定融合蛋白的表达和纯度.实验结果表明,成功地构建了NF-κB相互作用多肽的GST/多肽融合蛋白原核表达载体,进行了GST/多肽融合蛋白的诱导表达,融合蛋白的表达为可溶性表达,最终纯化获得纯度约为80%的可溶性GST/多肽融合蛋白,这将为后继利用GST pull-down,Bio-sensor,EMSA等实验进一步验证NF-κB相互作用多肽的功能提供可靠的材料来源奠定基础.  相似文献

构建融合重组表达载体并对其进行诱导表达蛋白以获得大量重组融合蛋白。通过PCR方法扩增出小鼠SDF-1α基因,克隆入pMD18T载体中,进行测序分析。将该基因亚克隆入原核表达载体pGEX-4T11中GST的下游构建重组质粒pGEX-4T1-SDF-1α,转化受态细胞BL21中,经IPTG诱导表达重组融合蛋白,对表达产物进行SDS-PAGE电泳检测分析。在约Mr36×103处出现一新生的蛋白条带。经灰度扫描检测,表达量约占菌体总蛋白的30%,纯化后得到了目的蛋白。成功克隆了小鼠的SDF-1α基因,并纯化融合基因GST-SDF-1α的原核表达产物。  相似文献

采用RT-PCR技术从番茄中扩增谷光甘肽转移酶基因ShGSTU1,构建该基因的原核表达载体pET28aShGSTU1和pET32a-ShGSTU1,并对重组质粒在大肠杆菌BL21(DE3)中表达条件进行优化,主要对诱导温度、诱导时间及诱导剂浓度进行分析.结果表明:重组蛋白在表达载体pET28a中的最佳表达条件为1mmol/L IPTG,37℃诱导4h;在表达载体pET32a中的最佳表达条件为1mmol/L IPTG,30℃诱导5h.  相似文献

目的 分析长爪沙鼠半胱氨酸蛋白酶抑制剂C (Cystatin C,CST3)cDNA序列同源性,并建立CST3蛋白原核表达体系,为长爪沙鼠CST3抗体制备和后续基因功能研究奠定基础。方法 对长爪沙鼠Cst3 cDNA序列进行克隆、同源性分析及密码子优化;将优化后序列酶切连接到pET28a载体,完成重组CST3蛋白表达载体构建;将该载体转化到感受态细胞中,通过异丙基硫代半乳糖苷(Isopropyl β-D-Thiogalactoside,IPTG)诱导实现CST3蛋白的原核表达,并用SDS聚丙烯酰胺凝胶电泳(SDS-PAGE)和蛋白质印迹法(Western blotting)验证。结果 长爪沙鼠Cst3基因与人和小鼠Cst3基因的序列同源性较低。密码子优化后的长爪沙鼠Cst3表达序列插入到pET28a质粒中,获得了CST3蛋白表达载体。经1 mmol/L IPTG 37 ℃诱导12 h,可获得大量CST3蛋白。结论 成功地构建了长爪沙鼠CST3蛋白的体外表达体系。  相似文献

克隆细粒棘球绦虫EgA31抗原基因,构建pET30a-EgA31原核表达载体,并在大肠埃希菌宿主系统中表达EgA31重组蛋白,对表达产物进行SDS-PAGE分析.从细粒棘球绦虫成虫组织中提取总RNA,反转录生成cDNA,以此cDNA为模板RT-PCR 克隆获得EgA31抗原基因,将其克隆至pUCm-T载体,测序确定其正确性.利用定向克隆技术将EgA31抗原基因片段克隆至原核表达质粒pET30a上,转化E coli DH5α,根据选择标记的卡那霉素抗性基因筛选到阳性克隆,通过PCR分析和酶切鉴定筛选出阳性克隆.IPTG初步诱导和表达pET30a-EgA31重组蛋白,SDS-PAGE电泳检测,并经凝胶图像分析确定目的蛋白的表达水平.测序表明选取的pET30a-EgA31阳性克隆均为正确连接EgA31抗原基因的重组质粒.经IPTG诱导后重组蛋白得到成功表达,在相对分子量约为31 kDa处有表达条带,表达量约占菌体总蛋白质的26%.成功克隆并构建了pET30a-EgA31原核表达质粒.初步诱导表达出EgA31重组蛋白,为进一步研究其免疫特性奠定了基础.  相似文献

利用RT-PCR技术从小鼠脂肪组织中扩增了OB基因,并利用定向克隆技术克隆至原核表达载体pTO-T7.将构建好的质粒导入表达型大肠杆菌BL21(DE3),IPTG诱导表达,并对表达产物进行Western-blot检测.表达产物利用亲和层析技术进行纯化.结果显示:表达的带有his标签的小鼠leptin融合蛋白,分子量约为20 ku.0.5 mmol/L的IPTG在37℃诱导2.5 h时,leptin融合蛋白的表达量达到最大,达菌体总蛋白50%以上.表达产物经过纯化,纯度可达90%以上.Western-blot杂交显示,小鼠leptin融合蛋白有很强的抗原特异性.  相似文献

通过聚合酶链式反应(PCR),扩增出交链孢菌激活蛋白辅助蛋白基因p42A.并将该基因按正确阅读框克隆到表达载体pGEx—KG中谷胱甘肽转移酶(GST)基因的下游。重组质粒转化大肠杆菌BL21,通过建立重组菌生长时间与OD600值的关系,及对菌液密度、诱导剂浓度、诱导时间和温度等条件的摸索,根据SDS—PAGE电泳检测结果判断融合蛋白的最佳表达条件。实验结果表明,最适诱导时期为重组菌生长对数中期;IPTG的最佳浓度为1.0mmol/L:37℃下诱导培养4h时产物表达量最高。超声波破碎菌体细胞,离心及电泳结果表明,表达产物为可溶性蛋白。  相似文献

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

以AC-13级配为基础,将橡胶颗粒代替部分集料掺入混合料中,以低温弯曲试验为评价方法对不同橡胶颗粒掺量下沥青混合料的低温抗裂性进行研究,并引入应变能密度值对混合料的低温抗裂性进行综合评价.试验结果表明:橡胶颗粒沥青混合料试件的破坏微应变均超过2 300,满足冬寒区的技术指标;无论是否掺加橡胶颗粒,随着温度的下降,沥青混合料破坏时的最大弯拉强度增大,弯拉应变降低,劲度模量增大;弯曲应变能密度在胶粒掺量为1%左右时具有较大的弯曲应变能密度值,此时橡胶颗粒沥青混合料具有较好的低温抗裂性.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
