Objective: To investigate the relationships between endothelial nitric oxide synthases (eNOS) G894T and 27 bp-variable number tandem repeat (VNTR) gene polymorphisms and osteoporosis in the postmenopausal women of Chinese Han nationality. Methods: In the present study, 281 postmenopausal women from Xi’an urban area in West China were recruited, and divided into osteoporosis, osteopenia, and normal groups according to the diagnostic criteria of osteoporosis proposed by World Health Organization (WHO). The bone mineral density (BMD) values of lumbar vertebrae and left hips were determined by QDR-2000 dual energy X-ray absorptiometry. Blood samples were tested for plasma biochemical indicators including testosterone, estradiol, calcitonin, osteocalcin, and procollagen type I amino-terminal propeptide by enzyme-linked immunosorbent assay (ELISA), tartrate-resistant acid phosphatase by spectrophotometric method, and the content of nitric oxide by Griess method. Genome DNA was extracted from whole blood, and G894T polymorphism of eNOS gene was analyzed by using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method and 27 bp-VNTR polymorphism of eNOS gene was genotyped by PCR method. Then the relationships between genotypes and biochemical indicators, genotypes and osteoporosis, and haplotypes and osteoporosis were analyzed. Results: The average BMD values of the femoral neck, ward’s triangle and lumbar vertebrae 1~4 (L1~L4) in the subjects with T/T genotype in eNOS G894T locus were significantly higher than those in the subjects with G/T and G/G genotypes (P<0.05). The average BMD of the femoral neck in the subjects with a/a genotype of eNOS 27 bp-VNTR locus was evidently higher than that in the subjects with b/b genotype (P<0.05). The plasma testosterone and osteocalcin concentrations in the subjects of eNOS G894T G/T genotype were evidently higher than those in the subjects of other genotypes (P<0.05); the plasma estradiol concentration in the subjects of eNOS 27 bp-VNTR a/a genotype was obviously higher than that in the subjects of b/b genotype (P<0.01). eNOS G/G homozygous frequencies in osteoporosis women, osteopenia women, and normal women were 85.37%, 76.38%, and 83.87%, respectively (P>0.05). 0% osteoporosis woman, 0.79% osteopenia women, and 3.23% normal women were eNOS a/a homozygous (P<0.05). The frequencies of eNOS 27 bp-VNTR a allele were 5.33% in the osteoporosis group, 10.24% in the osteopenia group, and 16.13% in the normal group (P<0.05, odds ratio (OR)=0.29, 95% confidence interval (CI)=0.11~0.77), suggesting that a/a genotype and a allele might have protective effects on osteoporosis. The haplotype analysis showed that G-b was 87.7% (214/244) in the osteoporosis group (P<0.05, OR=2.48, 95% CI=1.18~5.18). G-a was 5.3% (13/244) in the osteoporosis group (P<0.05, OR=0.29, 95% CI=0.11~0.77). G-b was a risk factor for osteoporosis, and G-a a protective factor. Conclusion: eNOS G894T G/T genotype influenced the plasma testosterone and osteocalcin concentrations, and T/T genotype influenced BMD. eNOS 27 bp-VNTR a/a genotype increased plasma estradiol concentration to have a protective effect on osteoporosis.  相似文献

目的观察帕米磷酸二钠防治绝经后骨质疏松症患者的疗效。方法将63例绝经后骨质疏松症患者随机分为两组,对照组采用口服凯思立D、罗盖全和仙灵骨葆胶囊治疗,治疗组采用静滴帕米磷酸二钠和口服凯思立D、罗盖全、仙灵骨葆胶囊治疗,分别治疗6个月。在治疗前、治疗1个月、3个月、6个月分别进行VAS疼痛评分,以及治疗前、治疗6个月进行腰椎L1-4骨密度(BMD)检测。结果治疗1个月和3个月,两组VAS评分较治疗前有不同程度降低,且治疗组止痛效果优于对照组(P0.05);治疗6个月,两组VAS评分均明显降低,但两组VAS评分无统计学差异(P0.05)。治疗6个月,两组腰椎L1-4骨密度较治疗前明显提高(P0.05),且治疗组优于对照组(P0.05)。结论帕米磷酸二钠可有效缓解绝经后骨质疏松症患者的疼痛,并且能提高患者的骨密度。  相似文献

Objective: To observe the effect of angiotensin II (Ang II) on expression of gap junction channel protein connexin 43 (Cx43) in the proliferation process of vascular smooth muscle cells (VSMCs) during the early stage of arteriosclerosis. Methods: Thirty-two adult male rabbits were randomly divided into 4 groups. Rabbits in Group A were fed common diet while others in Groups B, C, and D were fed high-cholesterol diet. Losartan (10 mg/(kg·d)) and ramipril (0.5 mg/(kg·d)) were added in the diet of Groups C and D, respectively. The animals were sacrificed after 8 weeks and abdominal aortas were removed and dissected. The expression of Cx43 was assayed using RT-PCR and Western Blotting analysis. Results: Cx43 was increased markedly in both protein and mRNA level in Groups B, C, and D fed high-cholesterol diet compared with that in control group (P<0.01). Cx43 level in losartan or ramipril treated groups was higher than that in control group (P<0.01, P<0.05), but lower than that in high-cholesterol diet groups (P<0.05, P<0.01). Conclusion: Cx43 level was upregulated in VSMCs during early atherosclerosis. Losartan and ramipril can inhibit the expression of Cx43.  相似文献

目的探讨2型糖尿病患者中亚临临甲状腺机能减退症与骨密度关系。方法选择32例2型糖尿病合并亚临临甲状腺功能减退患者作为观察组,36例甲状腺功能正常的2型糖尿病患者作为对照组,比较两组间腰椎1、2、3、4椎体和左侧股骨颈等部位的骨密度(BMD)。结果两组间在年龄、性别、BMI、糖尿病病程、糖化血红蛋白、FT3、FT4均无明显差异,观察组中TC、TG、LDL-C、HOMA-IR、TSH高于对照组,而HDL-C低于对照组,观察组左侧股骨颈BMD低于对照组,差异有统计学意义,两组间腰椎1、2、3、4椎体BMD无明显差异。结论 2型糖尿病合并亚临临甲减者存在股骨颈的骨密度降低,胰岛素抵抗明显,同时存在血脂代谢紊乱。  相似文献

This study aimed to observe the osseointegration of hollow porous titanium prostheses (HPTP) loaded with cancellous bone matrix (CBM) in rabbits using histological and biomechanical perspectives.Experimental samples were implanted into the lateral femoral condyles of 66 New Zealand rabbits,allocated into the following groups:non-porous prosthesis group (Group A,n=22);HPTP group (Group B,n=22);HPTP+CBM group (Group C,n=22).The rabbits were sacrificed at 3,8 and 12 weeks,postoperatively.X-ray analyses,microscopy techniques and morphological measurement software and mechanical tests were used for evaluation.At each time point,the tissues surrounding the implants were similar in all of the groups,with bony in-growth into the 2-mm round holes observed for the defects in Groups B and C.However,the internal bone formation was significantly better in Group C than in Group B at different time points (P<0.01).Biomechanically,the pull-out forces were significantly greater in Groups B and C than in Group A (P<0.01),with no difference between Groups B and C (P>0.05).These results suggest that bone can grow into the cavities of HPTP to achieve more stable locking fixation,and those osteogenic materials,such as CBM,can enhance osteogenesis to achieve better osseointegration between the implant and the host bone.  相似文献

The phyA^m gene encoding acid phytase and optimized neutral phytase phyCs gene were inserted into expression vector pPIC9K in correct orientation and transformed into Pichiapastoris in order to expand the pH profile ofphytase and decrease the cost of production. The fusion phytase phyA^m-phyCs gene was successfully overexpressed in P. pastoris as an active and extracellular phytase. The yield of total extracellular fusion phytase activity is (25.4±0.53) U/ml at the flask scale and (159.1±2.92) U/ml for high cell-density fermentation, respectively. Purified fusion phytase exhibits an optimal temperature at 55 ℃ and an optimal pH at 5.5-6.0 and its relative activity remains at a relatively high level of above 70% in the range ofpH 2.0 to 7.0. About 51% to 63% of its original activity remains after incubation at 75 ℃ to 95 ℃ for 10 min. Due to heavy glycosylation, the expressed fusion phytase shows a broad and diffuse band in SDS-PAGE (sodium dodecyl sulfate-polyacrylamide gel electrophoresis). After deglycosylation by endoglycosidase H (EndoHf), the enzyme has an apparent molecular size of 95 kDa. Objective: To study the relationship between plasma adiponectin concentration and the functional activities of circulating endothelial progenitor cells (EPCs) in patients with coronary artery disease (CAD). Methods: Circulating EPCs were enumerated as AC133+/KDR+ cells via flow cytometry and identified by co-staining with DiI-acLDL and fluorescein isothiocyanate (FITC)-conjugated lectin under a fluorescent microscope. The migratory capacity of EPCs was measured by modified Boyden chamber assay. Adhesion capacity was performed to count adherent cells after replating EPCs on six-well culture dishes coated with fibronectin. Results: The number of circulating EPCs (AC133+/KDR+ cells) decreased significantly in CAD patients, compared with control subjects [(74.2±12.3) vs (83.5±12.9) cells/ml blood, P<0.01]. In addition, the number of EPCs also decreased in CAD patients after ex vivo cultivation [(54.4±8.6) vs (71.9±11.6) EPCs/field, P<0.01]. Both circulating EPCs and differentiated EPCs were positively correlated with plasma adiponectin concentration. The functional activities of EPCs from CAD patients, such as migratory and adherent capacities, were also impaired, compared with control subjects, and positively correlated with plasma adiponectin concentration. Conclusion: The study demonstrates that the impairment of the number and functional activities of EPCs in CAD patients is correlated with their lower plasma adiponectin concentrations.  相似文献

Astragalus mongholicus (AM) derived from the dry root of Astragalus membranaceus Bge. var. mongolicus (Bge.) Hsiao is a widely used traditional Chinese medicine. The present study investigated the potential role of AM on renal fibrosis on a rat model of unilateral ureteral obstruction (UUO). We divided 48 Sprague-Dawley rats randomly into 4 groups: sham-operated group (Sham), untreated UUO group, AM-treated (10 g/(kg·d)) UUO group, and losartan-treated (20 mg/(kg·d)) UUO group as positive control. Haematoxylin & eosin (HE) and Masson staining were used to study the dynamic histological changes of the kidneys 7 and 14 d after operation. The expressions of fibronectin (FN), type I collagen (colI), hepatocyte growth factor (HGF), transforming growth factor-β1 (TGF-β1), and α-smooth muscle actin (α-SMA) were analyzed by real-time polymerase chain reaction (PCR), immunohistochemistry staining, and Western blot. Results show that, similar to losartan, AM alleviated the renal damage and decreased the deposition of FN and colI from UUO by reducing the expressions of TGF-β1 and α-SMA (P<0.05), whereas HGF increased greatly with AM treatment (P<0.05). Our findings reveal that AM could retard the progression of renal fibrosis. The renoprotective effect of AM might be related to inhibition of myofibroblast activation, inducing of HGF and reducing of TGF-β1 expression.  相似文献

目的探讨老年髋部骨折与髋部骨密度(BMD)和股骨近端几何结构的关系。方法老年髋部骨质疏松骨折47例为骨折组,无骨折50例为对照组,测量两组股骨近端骨密度及股骨颈轴长(FNAL)、颈干角(NSA)(髋部骨折组对其健侧进行检测),比较分析彼此间的相关性。结果老年髋部骨折组与对照组比较股骨近端骨密度明显降低,FNAL长于对照组,NSA较对照组大。结论股骨近端骨密度和FNAL及NSA可以提高对老年髋部骨质疏松骨折危险性的预测。  相似文献

Appressorium formation is an important event in establishing a successful interaction between the rice blast fungus, Magnaporthe oryzae, and its host plant, rice. An understanding of molecular events occurring in appressorium differentiation will give new strategies to control rice blast. A quick and reliable method to extract total RNA from appressorium is essential for studying gene expression during appressorium formation and its mechanism. We found that duplicate film is an efficient substratum for appressorium formation, even when inoculated with high density conidia. When inoculated with conidia at 1 × 106 ml^-1, the percentages of conidium germination and appressorium formation were (97.98±0.67)% and (97.88±0.45)%, respectively. We applied Trizol before appressorium collection for total RNA isolation, and as much as 113.6 lag total RNA was isolated from the mature appressoria at 24 h after inoculation. Functional analysis of two genes, MNH6 and MgATG1, isolated from the cDNA subtractive library, revealed that the quantity of RNA was good enough to construct a cDNA (complementary DNA) library or a cDNA subtractive library. This method may be also applicable for the appressorium RNA isolation of other pathogenic fungi in which conidia differentiate into appressoria in the early stages of host infection.  相似文献

目的探讨股骨近端骨密度与几何结构参数在老年人骨质疏松性髋部骨折诊治中的临床意义。方法将65例髋部骨折患者作为实验组。同期健康体检的60例老年志愿者为对照组。在骨盆片上测量股骨近端几何参数,在股骨颈、Ward’S三角和转子处测量BMD值(髋部骨折组对其健侧进行检测),对结果进行比较分析。结果实验组的BMD值低于对照组(P〈0.05);股骨颈骨皮质厚度与股骨颈BMD值有相关性(r=0.54,P〈0.05);股骨距内侧皮质厚度、转子处BMD值、颈干角和Ward’s三角BMD值相结合在预测髋部骨折中价值较大(r=0.78,r2=0.57,P〈0.05)。结论股骨近端骨密度结合其几何结构参数在骨质疏松性髋部骨折的诊治中具有较大的临床意义。  相似文献

目的 :比较 4种用于激素补充治疗的制剂的疗效和副作用 ,为选择用药提供参考。方法 :96例绝经妇女随机分为 4组 ,各服用克龄蒙、尼尔雌醇、妇复春和妇舒宁 6个月。用药前、后观察绝经期症状 ,检测血总胆固醇 (CHOL)、甘油三酯 (TG)、高密度脂蛋白 (HDL)和低密度脂蛋白(LDL)、骨密度及子宫内膜厚度。结果 :用药后 4组之绝经期症状均明显改善 ,Kupperman评分显著下降 ;除尼尔雌醇组TG下降及妇复春组HDL上升无统计学意义外 (P >0 0 5 ) ,其余各组CHOL、TG及LDL均明显下降 ,HDL明显上升 (P <0 0 5或 0 0 1) ;4组之骨密度均无明显变化 (P >0 0 5 ) ;无 1例宫内膜过度增生。对上述指标之影响 ,4种制剂间相比较无显著性差异 (P >0 0 5 )。结论 :4种制剂均能显著改善绝经期症状 ,有利血脂代谢和抑制骨丢失  相似文献

In order to develop a model for screening the agonists of human β2-adrenoceptor from Chinese medicinal herbs extracts, we used a cell-based functional assay based on a common G protein-coupled receptor (GPCR) regulation mechanism and destabilized enhanced green fluorescent protein (d2EGFP) reporter gene technique. The positive cell clone was confirmed by real-time polymerase chain reaction (PCR) and imaging analysis. To assess the value of this model, we screened over 2000 high performance liquid chromatography (HPLC)-fractionated samples from the ethanol extracts of Chinese medicinal herbs. Six fractions (isolated from Panax japonicus, Veratrum nigrum, Phellodendron amurense, Fructus Aurantii Immaturus, Chaenomeles speciosa, and Dictamnus dasycarpus) showed significant effects on active reporter gene expression, three of which (isolated from Phellodendron amurense, Fructus Aurantii Immaturus, and Chaenomeles speciosa) were selected for further concentration response analysis and the half maximal effective concentration (EC1/2 max) values were 4.2, 2.7, and 4.8 µg/ml, respectively. Therefore, this reporter gene assay was suitable for screening β2-adrenoceptor agonists. The results suggest that the six herbal extracts are the possible agonists of β2-adrenoceptor.  相似文献

We investigated the effects of fish protein hydrolysate (FPH) on growth performance and humoral immune response of the large yellow croaker (Pseudosciaena crocea R.). One thousand and two hundred large yellow croakers [initial average weight: (162.75±23.85) g] were divided into four groups and reared in floating sea cages (3 m×3 m×3 m). The animals were fed with 4 diets: basal diet only (control) or diets supplemented with 5%, 10% and 15% (w/w) FPH. The results show that dietary FPH levels significantly influenced the growth and immunity of the large yellow croaker. Compared with the control group, total weight gain (TWG) in all treatment groups, relative weight gain (RWG) and specific growth rate (SGR) in fish fed with diets supplemented with 10% and 15% FPH were significantly increased (P<0.05). Similar results were observed in immune parameters [lysozyme activity, serum complements, immunoglobulin M (IgM)]. Lysozyme activity, complement C4 and IgM were also significantly increased (P<0.05) in fish fed with diets supplemented with 10% and 15% FPH, while complement C3 level was significantly increased (P<0.05) in all treatment groups. In general, with the supplementation of FPH, particularly at dose of 10%, the growth performance and immunity of the large yellow croaker can be improved effectively.  相似文献

Objective: To study the relationships among magnetic resonance imaging (MRI), histological findings, and insulin-like growth factor-I (IGF-I) in steroid-induced osteonecrosis of the femoral head in rabbits. Methods: Thirty rabbits were randomly divided into experimental Group A (n=15) and control Group B (n=15). The 7.5 mg/kg (2 ml) of dexamethasone (DEX) and physiological saline (2 ml) were injected into the right gluteus medius muscle twice at one-week intervals in animals of Groups A and B, respectively. At 4, 8 and 16 weeks after obtaining an MRI, the rabbits were sacrificed and the femoral head from one side was removed for histological study of lacunae empty of osteocytes, subchondral vessels, and size of fat cells under microscopy, and the femoral head from the other side was removed for enzyme-linked immunoadsorbent assay (ELISA) for IGF-I. Results: At 4, 8 and 16 weeks after treatment, no necrotic lesions were detected in Group B, while they were detected in Group A. Light microscopy revealed that the fat cells of the marrow cavity were enlarged, subchondral vessels were evidently decreased, and empty bone lacunae were clearly increased. The IGF-I levels in Group A were significantly higher than those in Group B. At 8 weeks after the DEX injection, the MRI of all 20 femora showed an inhomogeneous, low signal intensity area in the femoral head, and at 16 weeks, the findings of all 10 femora showed a specific “line-like sign”. The MRI findings of all femora in Group B were normal. Conclusion: MRI is a highly sensitive means of diagnosing early experimental osteonecrosis of the femoral head. However, the abnormal marrow tissues appeared later than 4 weeks when the expression of IGF-I increased. This reparative factor has an early and important role in response to steroid-induced osteonecrosis of the femoral head, and provides a theoretical foundation for understanding the pathology and designing new therapies.  相似文献

To shorten operation time and improve survival rate of rats with myocardial ischemia or myocardial infarction, we use a novel device comprised of a face mask and a head/neck retainer in this study. We report the basic design of the novel respiratory face mask (RFM) and evaluate its performance in a rat model of myocardial ischemia. The device is cost-effective and easier to handle than other devices, such as tracheal intubation. Compared with conventional tracheal intubation, we found that RFM shortens operation time significantly while keeping blood indices normal; the mean operation time for rats in the mask group was (32±3) min, and that for the intubation group was (45±7) min (P<0.05). Moreover, the size and shape of the RFM can be changed according to the body weight of rats. In conclusion, RFM is an appropriate device for the establishment of myocardial infarction or ischemia-reperfusion in rats.  相似文献

目的:采用双能X线吸收法(DXA)进行大鼠骨密度(BMD)和身体软组织成分测量的精密度试验,探讨DXA对大鼠骨密度和身体软组织成分测量结果的可靠性。方法:对30只SD大鼠进行了DXA二次重复扫描,采用小动物专用软件分别分析大鼠上肢、下肢、躯干、全身等部位的骨密度,以及脂肪(fat)组织、肌肉(lean)组织和fat百分比,计算其精密度(以变异系数的均方根RMS-CV表示)。结果:SD大鼠全身骨密度测量精密度(RMS-CV)为0.9%,最小显著变化值为0.003 g/cm2;大鼠全身软组织成分:脂肪、肌肉组织的含量和fat组织所占软组织的百分比的精密度分别为9.6%、1.3%和9.7%。结论:DXA能较准确测量大鼠全身骨密度和软组织成分。  相似文献

Objective: To evaluate the effects oftramadol on the proinflammatory responses in a rat model of incisional pain by investigating its effects on nociceptive thresholds and serum interleukin-6 (IL-6) and IL-2 levels. Methods: Forty-two male Sprague-Dawley (SD) rats scheduled for plantar incision were randomly divided into 7 groups 01=6 in each group). Rats in Group 1 receiving general anesthesia with no incision were served as control; At 30 min before skin incision, Groups 2-5 were given 5 ml normal saline or 1, 10, and 20 mg/kg tramadol, respectively, intraperitoneally (i.p.); Group 6 received 10 mg/kg tramadol after operation; Group 7 received 10 mg/kg tramadol before incision, followed by 200 μg/kg naloxone after operation. Mechanical allodynia was measured by electronic yon Frey filament to evaluate the nociceptive thresholds 1 h before incision, and 1 h and 2 h after operation. Serum IL-6 and IL-2 levels were measured by enzyme-linked immunosorbent assay (ELISA) 2 h after operation. Results: Mechanical thresholds decreased significantly and serum IL-6 level increased significantly after operation in Group 2 compared with control (P〈0.01), and these changes were reversed respectively by tramadol in a dose-dependent manner (P〈0.05 and P〈0.01, respectively). IL-2 level remained unchanged after operation in Group 2, but decreased in Group 3 (P〈0.05), then gradually returned to the normal level in Groups 4 and 5. The intraperitoneally injected tramadol (10 and 20 mg/kg) produced a potent and dose-dependent antinocicptive effect on the lesioned paw. The antinocicptive effects of tramadol were partially an- tagonized by naloxone (200 μg/kg), suggesting an additional non-opioid mechanism. Conclusion: The results suggest that tramadol could be a good choice for the treatment of pain under the conditions that immunosuppression may be particularly contraindicated.  相似文献

为探讨牦牛骨粉对运动员骨密度、骨代谢和身体机能水平的影响,以击剑、现代五项、武术、拳击等项目45名运动员为对象,运动员每日服用5 g牦牛骨粉,每周6次,共4周。采用EXA-3000外周双能X线骨密度仪检测左右前臂、跟骨骨密度,抽静脉血检测骨代谢和身体机能相关生化指标。结果发现,服用牦牛骨粉后男子运动员右跟骨骨密度显著升高,女子运动员左右前臂、跟骨密度显著升高,服用牦牛骨粉后女运动员血清钙、磷和男运动员血清钙显著性降低,女运动员血清肌酸激酶显著性降低,男运动员IgM显著性升高。可见,服用4周牦牛骨粉对女运动员前臂、跟骨的骨密度有一定的改善作用,对男运动员前臂和跟骨的骨密度影响作用较小,对运动员骨代谢血液生化指标和免疫球蛋白无不利影响,可以较好地维持运动员的身体机能水平。  相似文献

为了研究抗阻力练习与人体骨健康状况与骨代谢之间的关系,分析抗阻力练习对人体骨健康状况与骨代谢的影响。在某大型健身房内随机选择40名成年人,随机将其划分成抗阻力练习组和对照组,每组20人。研究发现,经抗阻力练习后,人体的骨密度、骨矿含量、投影面积等均向有利于骨健康状况的方向发展,人体健康情况得到了一定程度的改善。说明抗阻力练习可促进骨代谢,对提高人体骨健康状况有积极的意义。为预防骨科疾病提供了可靠性依据。  相似文献
