携带p53基因重组腺病毒载体的构建

将pCMVp53重组转移载体经BamHI和NheI酶切,得到p53基因cDNA,然后将cDNA片段克隆到转移载体pCMV5GFP,使其受CMV5启动子的调控,获得pCMV5p53重组转移载体.用该线状重组转移载体与腺病毒右臂DNA经磷酸钙共转染293细胞获得重组腺病毒,经酶联免疫吸附法(ELISA)测定p53蛋白含量,证明外源p53基因在含重组腺病毒的293细胞中得到表达.     

携带p53和p21基因腺病毒转移载体的构建

构建携带p53和p21基因的重组腺病毒转移载体以研究p53和p21联合基因治疗效果.将p53cDNA克隆到转移载体pCMV5GFP替代gfp,得到pCMVp53,使其受到CMV启动子的调控;同时将p21基因克隆到pCMVp53,得到重组腺病毒转移载体pCMVp53/p21.得到携带p53和p21基因的重组腺病毒转移载体.     

基于植物p53保守序列构建RNAi的DNA片段方法

构建p53基因RNA干涉DNA片段的目的,是将其应用于植物悬浮培养中的细胞周期调节.依据拟南芥p53基因与其他高等植物具有高度保守区域的特点,合成其两个区域的核苷酸片段(Ⅰ和Ⅱ)及其相应的反向片段(Ⅰ′和Ⅱ′),并在Ⅱ和Ⅱ′端部分别引入内含子两个端部核苷酸序列.在相互连接后进行PCR选择扩增,其产物再与克隆载体连接并经过蓝白斑筛选获得重组DNA;在电泳和核苷酸测序鉴定后表明,最终得到了Ⅰ–5′–内含子–Ⅱ′–Ⅱ–内含子–3′–Ⅰ′序列的重组DNA片段.该片段两端含多克隆位点,通过插入植物的表达载体进入细胞基因组,在细胞中其转录产物将形成发夹结构,经胞内酶切后可以形成短的双链RNA片段,将具有干涉p53基因表达的功能.     

谷胱甘肽合成酶系基因协调表达体系的构建

通过 PCR获得 E.coli B谷胱甘肽合成酶系基因 ( gsh I,gsh II)片段 ,结合定点突变稀有起始密码子 ,设定 gsh I与 gsh II位置与距离 ,构建双顺反子重组表达载体 p Trc99A/gsh I- gsh II,建立GSHI、GSH- II蛋白表达体系。结果表明 :以 0 .0 8mmol/L IPTG于 2 8℃诱导工程菌 E.coli BL2 1( DE3) ( p Trc99A/gsh I- gsh II) ,GSH- I、GSH- II蛋白比为 4.5∶ 1 ( m∶ m)时谷胱甘肽合成能力最高 ,达到每克湿菌体 8.5 mg。通过构建单顺反子重组表达载体 p Trc99A/gsh I、p Trc99A/gsh II测定GSH- I与 GSH- II蛋白的最适配比 ,结果表明 :在 GSH- I、GSH- II蛋白总量恒定的情况下 ,要提高谷胱甘肽产率 ,两者比例以 3∶ 1～ 6∶ 1为宜     

人全长Notch1基因的克隆及其在HEK293 T细胞中的表达

目的:克隆人全长Notch1基因,构建人全长Notch1/p CMV6-Entry真核重组质粒并在HEK293 T细胞中进行瞬时表达.方法:采用PCR公知不认方法扩增人全长Notch1基因并克隆到真核表达载体p CMV6-Entry中并用限制性内切酶分析和DNA测序鉴定重组质粒.将该重组质粒转染HEK293 T细胞24h后,通过实时荧光定量PCR(q-PCR)和Western blot检测鉴定人全长Notch1的表达.结果:成功构建了人Notch1/p CMV6-Entry重组真核表达质粒,人全长Notch1基因在mRNA和蛋白水平上的表达显著高于p CMV6-Entry载体对照组和HEK293 T细胞对照组.结论:克隆获得人全长Notch1基因并验证在HEK293 T细胞中表达.     

大黄鱼与美国红鱼抗菌肽Piscidin串联基因的构建、酵母表达及抗菌活性鉴定

采用Piscidin基因串联表达的方案,通过Eco31Ⅰ限制性内切酶定向连接方法合成Pseudosciaena crocea-Sciaenops ocellatus-piscidin (PSP)串联基因.以毕赤酵母(Pichia pastoris)的穿梭表达载体pPICZαA构建重组表达质粒pPICZαA-PSP,转化入毕赤酵母SMD1168菌株中.应用实时荧光定量PCR方法进行多拷贝克隆筛选,实现了PSP重组串联肽的诱导表达和纯化,并对其抑菌活性进行初步鉴定,为piscidin抗菌肽的生产应用奠定基础.     

表达HIV-1复合多表位基因的p815细胞稳定株的建立和评价

建立稳定表达HIV-1p24MEG复合多表位基因的p815细胞克隆.设计引物,以HIV-1标准株全长cDNA序列为模板,PCR扩增获得p24基因片段;合成改造后的多个表位基因MEG并且与p24片段相连接,克隆入pcDNA3.1(+).在阳离子聚合物作用下重组真核质粒转染的p815(H-2d)细胞, 以G418压力筛选,RT-PCR检测mRNA表达,间接免疫荧光检测蛋白表达.通过PCR获得了HIV-1 p24片段,获得了与多表位基因连接后的p24MEG融合基因,成功构建了p24MEG基因的重组真核表达载体pcDNA3.1(+)/p24MEG.转染p815细胞后, RT-PCR检测到融合蛋白mRNA表达,间接免疫荧光显示 p815细胞内有融合蛋白的表达.结论:建立了稳定表达HIV-1p24MEG融合蛋白的p815细胞克隆,为评价多表位抗原p24MEG在BALB/c小鼠体内诱导的细胞免疫应答奠定基础.     

基于报告基因能筛选下调p66shc基因转录的药物普筛系统的建立

将p66shc基因的启动子序列克隆到含有荧光素酶报告基因和新霉素抗性基因的pGL3B-neo载体中,然后将重组质粒pGL3B-neo-p66shc转染入人的肺癌细胞A549中,通过单集落分离法得到含有重组质粒的稳定细胞系A549/p66shc。利用这个以报告基因为基础的药物普筛系统,筛选了数百种中药提取物及单体化合物,发现抑制率达到50%以上的2种中药提取物和8种单体化合物。     

紫球藻藻红蛋白α、β亚基在毕赤酵母和大肠杆菌中的表达

将紫球藻藻红蛋白α、β亚基基因(pe A、pe B)分别插入到毕赤酵母表达载体p AO815和大肠杆菌表达载体p ET-28a中.重组的共表达载体p AO815-pe A-pe B转化毕赤酵母(Pichia pastoris)GS115,经甲醇诱导、SDS-PAGE电泳及DOT-BLOT分析,发现藻红蛋白表达量低.p ET28a-pe A、p ET28a-pe B表达载体分别转入E.coli BL21,经IPTG诱导,Western Blotting分析,结果显示在约20.1 ku处有特异性蛋白条带,与融合蛋白(Pea-his、Peb-his)预期大小基本一致.     

一种与胆固醇7,8位脱氢相关蛋白的表达研究

家蚕(Bombyx mori)的Nvd-Bm蛋白是一种与胆固醇7,8–脱氢酶相关的蛋白.根据NCBI中报道的家蚕中的胆固醇7,8–脱氢酶基因序列(Gen Bank登录号:AB232986.1),采用密码子优化及基因克隆技术,扩增获得胆固醇7,8–脱氢酶目标基因nvd-Bm,并构建了重组穿梭载体p PIC9K-nvd-Bm,通过电转化,成功构建了含有p PIC9K-nvd-Bm的毕赤酵母(Pichia pastoris)GS115异源真核表达工程菌株.经SDS-PAGE蛋白电泳分析表明,含有p PIC9K-nvd-Bm的毕赤酵母GS115表达菌株能够成功表达目标蛋白,为胆固醇的7,8–脱氢产物即7–脱氢胆固醇的生物转化奠定了基础.     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.     

Lower Paleozoic oil relationships within Williston Basin, Canada

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.     

An Analysis of Catherine＇s Character——Wuthering Heights

Characterization of Ordovician carbonate reservoirs, southeastern Saskatchewan, Canada

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3～4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰～ -7.8‰, PDB) and matrix dolomites (-6.6‰～ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084～0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078～0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.

Design and analysis of generic LBS application developing platform

Location based services is promising due to its novel working style and contents.A software platform is proposed to provide application programs of typical location based services and support new applications developing efficiently. The analysis shows that this scheme is easy implemented, low cost and adapt to all kinds of mobile nework system.     

橡胶颗粒沥青混合料低温抗裂性试验研究

以AC-13级配为基础,将橡胶颗粒代替部分集料掺入混合料中,以低温弯曲试验为评价方法对不同橡胶颗粒掺量下沥青混合料的低温抗裂性进行研究,并引入应变能密度值对混合料的低温抗裂性进行综合评价.试验结果表明:橡胶颗粒沥青混合料试件的破坏微应变均超过2 300,满足冬寒区的技术指标;无论是否掺加橡胶颗粒,随着温度的下降,沥青混合料破坏时的最大弯拉强度增大,弯拉应变降低,劲度模量增大;弯曲应变能密度在胶粒掺量为1%左右时具有较大的弯曲应变能密度值,此时橡胶颗粒沥青混合料具有较好的低温抗裂性.

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
