离心淘洗技术在药物细胞毒性研究中的应用

应用离心淘洗技术同步分离体外培养小鼠红白血病细胞,得到同步化的G_1期,S期和G_2M期细胞。经药物处理后进行细胞集落形成实验,发现钙拮抗刺异博定对各时相细胞集落形成率影响很小。而抗癌药物平阳霉素对各时相细胞都有较强毒性,特别是对G_2M期细胞,杀伤作用特别强。

USE OF CENTRIFUGAL ELUTRIATION IN STUDIES ON CYTOTOXICITY OF DRUGS

The synchronized G_1, S, G_2M phase cells of cultured MELC celts were acquired by Centrifugal Elutriation. It was discovered that survival fractions of the synchronized cells treated with a calcium antagonist verapamil were similar with those controlled in colony forming tests. The cytotoxicity of antitumor drug pingyangmycin to the synchronized cells was higher, especially to the G_2M phase cells.
