脱水胰凝乳蛋白酶制备方法的改进

摘    要:对脱水胰凝乳蛋白酶(AnCht)的制备方法进行改进.在粗品AnCht亲和层析纯化前再进行一次PMSF化学修饰,降低AnCht中残存的蛋白水解酶活力.分析表明,用该方法制得的AnCht与抑制剂BPTI结合的化学计量及强度均接近于理论值,并且其催化活力几乎全部丧失,表明纯度很高,可用于Cht与其底物间的分子识别

Abstract:We improved the usual method for the preparation of anhydrochymotrypsin (AnCht). Before the crude AnCht was purified by affinity chromatography, it was chemically modified by PMSF once again. This strategy greatly reduced the residual proteinase activity of the product. The further analysis demonstrated that the product can bind to chymotrypsin inhibitor BPTI stoichiometrically and the purity of it is high. Therefore, it can be used in the studies of molecular recognition between chymotrypsin and its substrates.
