摘    要:糖基磷脂酰肌醇(GPI)转酰胺基酶复合物将GPI脂连接到新合成的蛋白质上,在酵母中包括GAA1,GPI8,GPI16,GPI17,CDC91五种亚基,皆为必需基因.采用含TAP标记转酰胺基酶亚基的酵母细胞株,利用TAP法在自然状态下提取、纯化分离蛋白复合物的优势,鉴定与靶蛋白相关联的蛋白.对Gp i8p,Gp i16p,Gp i17p进行了研究.实验结果表明,酵母只含一个拷贝的Gp i8p,Gp i16p,Gp i17p亚基,为进一步研究转酰胺基酶复合物的功能和作用机制提供了线索.

Abstract:Glycosylphosphatidylinositol(GPI) transamidase complex transfers GPIs to newly synthesized proteins.Genetic and biochemical approaches have so far identified 5 proteins that compose transamidase subunits.All the five proteins are essential in yeast.To illuminate the role of yeast transamidase subunits and to reveal the structure-function relationships in this complex,the complex was extracted and purified from yeast corresponding strains harboring TAP tagged transamidase subunit,the proteins'interaction with the target subunits under native condition was identified.The results show that the yeast transamidase complex only contains a single copy subunit of Gpi8p,Gpi16p and Gpi17p.
