连续灌注培养红豆杉细胞生产紫杉醇的研究

摘    要:利用由一圆形柱和三角瓶组成的生物反应器对红豆杉细胞进行连续灌注培养,结果显示,培养基的pH值,糖和磷的变化幅度较平缓,细胞的生长期长,说明该反应器适于细胞生长,根据该反应器具有培养液提取简便的特点,首次提出诱导子可按照培养基中残糖和残磷的体积质量加入,而且发现高含量紫杉醇仅发生在特定的残糖和残磷体积质量下,结果还显示该反应器用于诱导和萃取结合处理细胞,与搅拌式和气升式反应器相比,得到较高的生物量和紫杉醇产量,这些结果表明该反应器可用于连续灌注培养红豆杉细胞生长紫杉醇。

Abstract:Taxol production from Taxus chinensis cells in a continuous perfusion culture bioreactor consisted of a cylindrical column and a flask was studied. The results showed that the changes of pH, sugar and phosphate in the bioreactor were relatively slow, and growth phase was comparatively long, indicating that the bioreactor was suitable for cell growth. In view of that it was easy to sample the culture medium in this bioreactor, it was firstly proposed that elicitors could be added according to the concentrations of sugar and phosphate in the medium, and observed that higher taxol production only appeared at certain concentrations of sugar and phosphate. It was also demonstrated that combined treatment of elicitation and extraction on Taxus cells in the bioreactor achieved higher biomass and taxol yield than those in the stirred bioreactor and airlift bioreactor. The data implicated that the bioreactor was available for producing taxol by continuous perfusion culture.
