摘    要:从构建的对虾白斑综合症病毒cDNA文库中,我们筛选到一个拷贝数最高,编码82个氨基酸的基因(命名为p9,wsv230).该基因被克隆到pGEX-2T载体中进行GST融合蛋白的原核表达,纯化的融合蛋白GST-P9用于制备特异的多克隆抗体.利用微阵列技术和免疫荧光标记技术,对该蛋白的转录水平及其在感染细胞内的分布进行了初步研究,表明该基因为高丰度表达,推测是对虾白斑综合症病毒的一个重要基因.

Abstract:White spot syndrome virus(WSSV) has been the major pathogen that led to severe mortalities of the cultured shrimps in recent years.An open reading frame(termed the p9 gene) encoding 82 amino acids was revealed,which was most abundant in the WSSV cDNA library.The gene was cloned into pGEX-2T and expressed as a fusion protein with glutathione S-transterase(GST).Specific antibody was raised by using the purified fusion protein(GST-p9).With the technique of microarray and immunofluorescent labeling,the gene expression level of p9 protein and its location in the infected cells were studied.The results showed that p9 gene was highly expressed and may be an important gene of WSSV.
