六种蝗虫基因组DNA多态性的RAPD标记研究

摘    要:运用随机扩增多态 DNA技术对斑翅蝗科 Oedipodidae中三属六种蝗虫基因组DNA进行了多态性比较 ,共扩增出 2 2条特异性片断 ,分子量大小为 650~ 1 990 bp之间 ,并根据各种间片段共享度构建了 UPG聚类关系图 .研究结果表明 ,大垫尖翅蝗和甘蒙尖翅蝗的片段共享度为 0 .375;红翅皱膝蝗和鼓翅皱膝蝗的片段共享度为 0 .2 86;亚洲小车蝗和黄胫小车蝗的片段共享度也为 0 .2 86

Abstract:In this paper is studied the polymorphism of genomic DNA in 6 species of locusts belonging to 3 genera of the family Oedipodidae by means of random amplified polymorphic DNA(RAPD).All in all,22 specific DNA segments were obtained by amplification with the primer OPA-02,with their molecular weight ranging from 650 bp to 1990 bp.Dendrogram of the 6 species based on UPGMA analysis demonstrated the gradually greater generic variation from species of the same genus to those belonging to a different genus,the average similarity being higher in Oedaleus and Epacromius genera than in Angaracris genus.Relationships among the 6 species were examined by comparison of the segments.Cluster analysis of the results is in compliance with findings from morphological classification.
