石蒜碱对人胃癌细胞体外增殖的影响

摘    要:以体外培养的h GCCs MGC-803细胞系为研究对象,采用不同浓度石蒜碱(5、2.5、1.25μmol/L)进行处理,倒置显微镜下观察其生长状态,同时采用血球计数板统计其体外增殖率,采用流式细胞技术检测处于不同细胞周期的细胞比例.结果表明,石蒜碱处理对h GCCs的体外增殖具有一定的抑制作用,其中5μmol/L石蒜碱对h GCCs的抑制作用最为明显.显微观察和计数分析结果表明,5μmol/L石蒜碱处理的h GCCs活力最低,细胞凋亡现象最明显.细胞周期检测的结果显示石蒜碱处理会使h GCCs增殖趋缓,细胞周期延长.

Abstract:The h GCCs were treated with different concentration of lycorisradiata alkali( 5、2. 5、1. 25 μmol / L respectively),growth status were observed by the inverted microscope,and proliferation rate were count by hemocytometer,the proportion of h GCCs in different cell cycles were detected by flow cytometry. The results showed that the proliferation of h GCCs was suppressed by treated with lycorisradiata alkali,the most obvious suppressed of h GCCs was 5μmol /L treatment group. Through microscopic observation and counting analysis results,it has been found that the vitality of h GCCs was the lowest and the cells apoptosis was the most obvious by treated of 5 μmol / L lycorisradiata alkali. The result of cell cycle detection revealed that by treated with lycorisradiata alkali,the reproduction of h GCCs became slow and cell cycle became extended.
