“索邦”百合茎尖培养及植株再生的研究

以“索邦”百合的小鳞茎作为外植体，分别对外植体的表面消毒、茎尖脱毒、小鳞茎增殖与生根培养等进行研究。结果表明：用酒精（30s）＋次氯酸钠（10min）＋升汞（6min）交替消毒，外植体存活率达68．1％；热水处理30min结合茎尖培养，脱毒效果显著，脱毒率可达80％；小鳞茎在6-BA0．5mg·L^-1＋IBA0．2mg·L^-1激素组合下，平均增殖系数达6．3；用1／2MS＋NAA0．6mg·L^-1培养基诱导生根，生根率100％，且根系粗壮。脱毒试管苗移栽应在防虫网室内进行，基质选用经过严格消毒的泥炭土和珍球岩混合基质（体积比为3：1），移栽成活率可达95％以上。

A STUDY ON SHOOT TIP CULTURE OF LILIUM ORENTIAL "SORBONNE" AND PLANT REGENERATION

As the explant, the bulblet section of Lilium orential ＂Sorbonne＂was studied to measure the treatment effects of surface sterilization, stem apex detoxifieation, bulblet proliferation and roofing culture, respectively. The results revealed that survival rate of explant reached 68.1% by the treatments of surface sterilization with 75% alcohol （0.5 minute） ,5% NaCIO solution （ 10 minutes） and 0.1% HgCl2 solution （6 minutes）. Due to the hot water wash for 15 minutes and shoot tip culture treatments, the stem apex detoxification rate reached to 80%. The average multiplication coefficient reached to 6.3 for explants at the improved culture medium of 6 - BA0.5mg · L^-1 ＋ IBA0.2mg · L^-1. The radication rate reached to 100% at the improved culture medium of 1/2MS ＋ NAA0.6mg · L^-1; and the roots were thick and strong. The transplant of virus free seedling should be carried out in the net insectary. The planting cultural mixture needed to be disinfected strictly. The mixture was made of peat soil and pearl rock powder （the physical volume compares to 3：1 ）. The transplant survival ratio could reach to 95% above.
