高原环境心肌细胞体外生长特性研究

摘    要:目的:研究高原环境心肌细胞体外生长特性,作为高原环境研究心肌细胞功能的实验基础。方法:将心肌细胞系复苏后使用体积分数为0.15的高糖DMEM培养基接种于33mm培养皿中并置于37℃5%CO2培养箱中进行培养;台盼蓝染色法检测细胞的成活率,并观察心肌细胞的形态变化,CCK8法绘制细胞生长曲线,血球计数法进行细胞计数。结果:心肌细胞最初接种时呈圆形,24小时后细胞贴壁生长;4天后细胞有伪足伸出,细胞呈梭形、多角形等形态,大小均匀,生长迅速,7天后细胞逐渐形成细胞簇,11天后细胞簇相互连接成片生长,达90%融合时进行传代;计数1000个细胞,蓝染细胞48个,成活率为95.20%;生长曲线显示细胞生长过程分为潜伏期、对数生长期和平台期三个阶段。结论:高原环境能够进行体外心肌细胞培养。

Abstract:The growth characteristics of myocardial cell in vitro was analyzed as an basic experiment for the re-search on myocardial cell function at high altitude. The recovered myocardial cell was cultured in 5% CO2 incu-bator at 37 ℃ by inoculating in petri dishes with high glucose DMEM medium containing 15% of FBS. Trypanblue staining was used to detect cell survival rate and morphological changes in myocardial cell. The growthcurve of myocardial cell was drawn by CCK8 method and the myocardial cell were counted by blood cell count-ing method. The results showed that the myocardial cells were rounded initially and it growing adherently after24 hours. The cells extended pseudopodia four days later and showed a rapid growth and uniform in size andspindle, polygonal shape morphologically and so on. Cells gradually formed cluster after 7 days, and the Cellclusters interconnected into 90% fusion in batches after 11 days. 1000 cells and 48 aizen cells were counted,and the survival rate reach to 95.20%. The cell growth curve showed three growing stages such as incubation pe-riod, logarithmic growth phase and platform growth phase. In conclusion, the myocardial cell can be cultured invitro at high altitude envrionment.
