| 摘 要： | 以兔骨胳肌为实验材料,构建了兔骨骼肌cDNA文库,根据该基因保守序列,设计简并引物,利用RT-PCR技术,克隆了兔MSTN基因EST片段,以EST片段为探针,应用Southern杂交技术筛选文库,克隆了兔肌肉生长抑制素基因全长cDNA并在GenBank注册(注册号:AY169410).用生物信息学方法对该基因进行了比较分析,表明从氨基酸序列及进化角度兔和其他哺乳类生物的肌肉生长抑制素基因之间关系密切.

| Abstract: | In this study, we construct cDNA library of rabbit in skeletal muscle. To clone the expressed sequence tags (ESTs) of rabbit myostatin, we designed degenerate oligonucleotides corresponding to conserved regions of myostatin and used these oligonucleotides as primers for RT-PCR. A full-length myostatin cDNA(NCBI GenBank:AY169410) clone was obtained from screening a rabbit skeletal muscle cDNA library by southern blot using ESTs as probe. To identify the full-length cDNA of myostatin, we compared the nucleotides sequences and amino acid sequences with other animals by bioinformatics. An alignment of the predicted amino acid sequences of the rabbit myostatin proteins with other animals exhibits 95% similarity. The predicted rabbit myostatin amino acid sequence contains all the hallmarks characteristic of members of the TGF-β superfamily, including a signal sequence for secretion, a proteolytic processing site, and a carboxy-terminal region containing the conserved pattern of cysteine residues. |
