重组人碱性成纤维细胞生长因子类似物在大肠杆菌中的分泌表达研究

用带有大肠杆菌外膜蛋白信号肽序列的重组分泌型表达载体pSE-hbFGF，在大肠杆菌BL21中分泌表达了rhbFGF类似物。SDS－PAGE、免疫印迹结果表明，表达产物分泌到了细胞周质和培养液中，用间接ELISA测定出培养上清中的rhbFGF类似物为24.5mg/L，周质中为36.5mg/L菌液（O.D600nm=10)。同时探讨了发酵培养中影响rhbFGF表达的几个关键因素，如诱导时期的选择、诱导时间的长短、培养基的选择及诱导剂的浓度等。

The Study on Secretory Expression of Recombinant Human bFGF Analog In E. coli

Human basic fibroblast growth factor (hbFGF) gene was expressed and secreted by recombinant expression vector pSE hbFGF with Omp A signal sequence in E.coli BL21(DE3pLys). SDS PAGE and Western blot analysis verified that the expressed rhbFGF analog was secreted into culture medium and periplasm of bacterial. The yields of rhbFGF anolog secreting into medium and periplasm are 24.5mg/L and 36.5mg/L (O.D 600nm =10) respectively by indirect ELISA.And also the key factors influncing fermentation culture has been studied.It has been showed that the choices of the induction period,induction time,culture medium and the inducer concentrations are very important.
