摘    要:利用人骨形成蛋白-4(hBMP4)与人骨形成蛋白-7(hBMP7)的成熟肽cDNA片段制备转基因毕赤酵母菌株,实现了hBMP4与hBMP7在巴斯德毕赤酵母细胞中的共表达.Western-blotting分析表明,表达产物含有hBMP4与hBMP7,片段大小分别为26 kD和17 kD,均为单体蛋白.

Abstract:The techniques for co-expression of the mature domain of hBMP4 and hBMP7 in Pichia pastorls were established. The cDNA mature domains that encodes BMP4 and BMP7 were cloned into pPICgK and the resulting constructs were transformed into Pichia pastoris for co-expression. Western-blotting analysis by anti-BMP4 antibody and anti-BMP7 antibody demonstrated that hBMP4 and hBMP7 were co-expressed in culture supernatant. Further analysis showed that the expressed hBMP4 and hBMP7 were monomers, with the molecular weight of 26 000 and 17 000 dalton, respectively.
