发酵过程中小单孢菌40027及其衍生菌株的抑菌作用

摘    要:研究了不同遗传背景下福堤霉素A的产生菌——小单孢菌40027菌株及整合了质粒pSET152菌株的抑菌作用.以金黄色葡萄球菌为指示菌跟踪不同时期小单孢菌40027菌株和Micromonospora sp. 40027::pSET152菌株的抑制活性.结果表明:小单孢菌40027菌株抑菌活性比Micromonospora sp .40027::pSET152的抑菌活性强;小单孢菌40027菌株和Micromonospora sp. 40027::pSET152菌株分别在种子液制备第1 d及第2 d时菌种产素能力较大.小单孢菌40027菌株和Micromonospora sp. 40027::pSET152菌株在发酵液制备第4 d为最佳产素时间.分别采用超声波法和反复冻融法破碎菌丝体,结果表明超声波破碎法较为理想,为进一步研究小单孢菌胞内产素情况打下了基础.

Abstract:The bacteriostasis of fortimicin A producing strains Micromonospora sp.40027 and Micromonospora sp.40027 with pSET152 were studied based on their different genetic backgrounds.Staphylococcus aureus is considered as an indicator to track bacteriostatic activity of Micromonospora sp.40027 and Micromonospora sp.40027::pSET152 strain.The results showed that Micromonospora sp.40027 strain has higher bacteriostatic activity than Micromonospora sp.40027::pSET152 strain.Their seed liquids have higher fortimicin.A production on the firist day and second days individually;Both Micromonospora sp.40027 and Micromonospora sp.40027::pSET152 strain have the highest fortimicin A production at the fourth day of fermentation culture.Mycelium was disrupted by ultrasonic method and freeze-thaw method.The results showed that ultrasonic method is better for mycelium disruption than freeze-thaw method.The study on using different methods to disrupting mycelium can be used for further investigation of Micromonospora intracellular antibiotics.
