内生真菌SF4的鉴定及产乙酰胆碱酯酶抑制剂发酵条件的初步优化

摘    要:基于菌株菌落和显微形态特征以及ITS-rDNA序列分析结果,将产乙酰胆碱酯酶抑制剂(AChEI)的内生真菌SF4鉴定为枝顶孢霉属Acremonium sp.真菌,命名为Acremonium sp.SF4.通过对菌株SF4发酵的基础培养基、发酵温度、发酵时间、摇床转速进行初步优化,确定了其最适产AChEI的摇瓶发酵条件为:沙氏培养基,发酵温度28 ℃,摇床转速为150 r·min-1,发酵培养14 d.在此条件下可发酵获得含AChEI的发酵浸膏0.331 g·L-1,对乙酰胆碱酯酶的抑制率可达60.27%,相比优化前均有提高.研究有助于后期规模化高效发酵,为分离、纯化和结构解析单一组分AChEI奠定基础.

Abstract:The acetylcholinesterase inhibiters(AChEI)-producing endophytic fungi strain SF4 was identified as Acremonium sp.SF4 based on its morphological characteristics and ITS-rDNA sequence analysis.The effects of fermentation medium,temperature,rotation speed and cultural time on acetylcholinesterase(AChE)inhibitory activity and AChEI metabolites amount were analyzed.It was indicated that the appropriate fermentation condition was:sabouraud dextrose broth(SDB)medium,28 ℃,150 r·min-1 for 14 d.Under the optimized conditions of fermentation,the amount of AChEI metabolites and AChE inhibitory activitycytotoxic activity is 0.331 g·L-1,and 60.27% respectively,higher than initial conditions.These results can lay the foundation for the further separation and purification of the AChEI metabolites.
