基于分子印迹膜的表面等离子体共振传感器检测黄体酮

摘    要:分别采用热聚合和光聚合的方法在金膜表面合成黄体酮分子印迹膜,并将其作为识别元件应用于表面等离子体共振传感器检测黄体酮. 通过对其进行洗脱及吸附研究发现:采用乙腈/乙酸混合液可以快速将黄体酮洗脱下来,洗脱前后共振角分别移动了1.15°和0.72°;该传感器实现对黄体酮的最低检测限均为10-14 g/mL;在10-14~10-6 g/mL的浓度范围内,共振角度的变化与黄体酮浓度的负对数成线性关系. 同时将具有黄体酮识别位点的分子印迹膜分别吸附浓度为10-4 g/mL的雌二醇,睾酮和睾酮丙酸酯溶液,结果显示均没有非特异性吸附作用. 以上结果表明,基于黄体酮分子印迹膜的表面等离子体共振传感器能够对黄体酮实现快速定量的专一性检测.

Abstract:A surface plasmon resonance (SPR) sensor based on molecularly imprinted film (MIF) was proposed for the sensitive detection of progesterone. The MIF was synthesized by the method of thermo and photo polymerization on the sensor surface, which was subsequently removed to form the recognition cavities by the mixture of acetonitrile and acetic acid. The SPR sensor develops the angle shift by 1.15° (thermo polymerization) and 0.72° (photo polymerization) after washing progesterone from MIF and allowed detection of progesterone at concentration down to 10-14 g/mL. The resonance angle changes show a good linearity versus minus logarithmic concentration of progesterone from 10-14 g/mL to 10-6 g/mL. A control experiment with the analogues estradiol, testosterone and testosterone propionate (10-4 g/mL) shows a good selectivity of the MIF for the recognition of progesterone.
