辛基酚下调AIB1、C-erbB2表达并降低乳腺癌发病的研究

摘    要:乳腺癌是女性最常见的恶性肿瘤,其危险因素与雌激素、环境因素改变和某些基因突变有关.研究探讨出生早期大鼠接触辛基酚(OP)对乳腺癌发病的影响及乳腺癌中AIB1、C-erbB2表达的影响.雌性SD大鼠随机分为对照组(Con)、模型组(Mod)、OP20组、OP40组、OP80组,Gen组.d22-d28皮下注射OP式Gen,d220处死大鼠.Con组未发生肿瘤,OP20、OP40、OP80、Gen和Mod组的乳腺癌发生率分别为57.14%、28.57%、52.38%、23.81%和66.67%;与Mod组相比,OP40组和Gen组大鼠乳腺癌组织中AIB1和C-erbB2mRNA明显降低并维持一定时间.研究提示OP40能降低DMBA诱导乳腺癌的发生,原因可能与下调乳腺癌中AIB1和C-erbB2表达有关.

Abstract:The breast cancer is the most common malignancy of women.This risk is concerned with estroGen,environment and some specific Genes mutations. This study is to investigate the effects of breast cancer incidence rate and AIB1,C-erbB2 expression in breast cancer tissue on early rats treated with environmental estroGenic compounds such as Octylphenol (OP) and Genistein (Gen). Female Sprague-Dawley rats were randomly divided into control group,model group(Mod),OP20,OP40,OP80,and Gen.Octylphenol was given to rats via subcutaneous injection from d22 to d28. DMBA was given to rats via oral gavage on d42. Rats were killed on d220. There was no breast cancer in Con group. The incidence rate of breast cancer in OP20 ,OP40 ,OP80,Gen and Mod was 57.14%、28.57%、52.38%、23.81% and 66.67% respectively. Compared with Mod,40mg/kg OP and Gen decreased AIB1 and C-erbB2 mRNA expression significantly in breast cancer and lasted for a long time. The study suggests that OP40 and Gen can decrease DMBA-induced breast cancers,which may be related to down-regulation of AIB1 and C-erbB2 expression in breast cancer.
