摘    要:选用大花萱草新品种"baltimore oriole”的叶片、根段、茎尖、花茎、花托、花瓣和子房7种外植体,对愈伤组织的诱导进行研究.结果表明:茎尖外植体诱导从生芽效果最好,不受时间限制,但是污染率很高,且损坏花苗;花茎、花托、花瓣和子房取材受时间限制,花茎的愈伤组织诱导率较高,花托、子房愈伤组织诱导率较低,花瓣难以诱导出愈伤组织;叶片和根段取材方便但是难以诱导出愈伤组织.从茎尖和花茎诱导出的丛生芽及愈伤组织,转入继代培养基后获得了新生芽,而花托和子房诱导的愈伤组织干枯死亡.因此,大花萱草较适宜的外植体材料为茎尖和花茎.

Abstract:Using the leaf,root,stem apex,scape,receptacle,petal and ovary of Hemerocallis hybrida as explants,callus and regenerated plant were studied.The results showed that stem apex was the most suitable explants which were induced to shoot clump.The stem apex had no time limits,but they were easily polluted and the plants were also damaged.Scape,receptacle,petal and ovary had time limits.The inductivity rate of callus of scape was higher than receptacle and ovary and petal couldn’t be induced.The leaf and root couldn’t also be induced although they could easily be obtained.The regenerated plants were obtained from callus and shoot clump that were from stem apex and scape.So the conclusion was that stem apex and scape were the more suitable explants.
