绞股蓝多糖对2型糖尿病大鼠血糖的影响及其机制初步研究

摘    要:目的:研究绞股蓝多糖对实验性2型糖尿病大鼠血糖、血脂的影响及其可能的机制。方法:采用Wistar大鼠高脂高糖饲料结合小剂量链脲佐菌素建立2型糖尿病大鼠模型,通过测定空腹血糖(FBG)和葡萄糖耐量,观察灌胃给予绞股蓝多糖对糖尿病大鼠的治疗作用,同时测定大鼠的血清胰岛素(Ins)、总胆固醇(TC)、甘油三脂(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)及部分抗氧化系统相关指标。结果:给予绞股蓝多糖后FBG明显降低,葡萄糖耐量曲线明显降低。此外,绞股蓝多糖可明显降低2型糖尿病大鼠的血清TC、TG、LDL和丙二醛水平,提高模型鼠的血清Ins、HDL、超氧化物歧化酶活性。结论:绞股蓝多糖对实验性糖尿病大鼠有明显的降血糖、降血脂作用,其作用可能与增加Ins水平,增强抗氧化能力有关。

Abstract:Objective: To investigate the hypoglycemic and hypolipidemic effects of polysaccharide from Gynostemma pentaphyllum on type 2 diabetic rats and discuss the mechanisms. Methods: Type 2 diabetes rat model was induced by injection of streptozotocin after feeding high-fat and high-sugar forage. The rats were divided into six groups. After the rats were treated with Gynostemma pentaphyllum polysaccharide or positive control agent for 2 months, the changes of blood glucose, insulin, total cholesterol, triglycerides, high density lipoprotein, low density lipoprotein and other biological indexes were assessed. Results: Administration of the polysaccharide from Gynostemma pentaphyllum reduced blood glucose in a dose-dependent manner and decreased glucose tolerance curve. Compared with model group, Gynostemma pentaphyllum polysaccharide groups significantly decreased blood total cholesterol, triglycerides, low density lipoprotein and malonaldehyde, and increased blood insulin, superoxide dismutase and high density lipoprotein. Conclusions: These data indicate that Gynostemma pentaphyllum polysaccharide has hypoglycemic and hypolipidemic effects in rats with STZ-induced type 2 diabetes. The mechanism may be related with the increasing of serum insulin and antioxidant activity.
