摘    要:建立了游离胶分馏器(OFFGEL)和反相液相色谱质谱(LC-MS)联用分析SH-SY5Y细胞分泌蛋白质组的新方法.收集SH-SY5Y胞外蛋白用胰酶酶切成肽段后,用OFFGEL进行预分离并用LC-MS进行检测.用Mascot检索出88个蛋白,再通过生物信息学软件SignalP对Mascot检索的蛋白进行预测和筛选,共得到31种蛋白,用SingalP中的神经网络法和隐马可夫模型对其中的CERU_HUMAN进行分析,发现该蛋白是分泌蛋白,且信号肽的剪切位点在第19和第20氨基酸之间,软件SingalP提升了鉴定分泌蛋白的可靠性.
