嗜热四膜虫接合后体发育期皮层细胞骨架蛋白组分的比较研究

摘    要:该文以嗜热四膜虫(Tetrahymena thermophila)BF株BF1、BF5系细胞为材料,采取生化抽提、SDS-聚丙烯酰胺凝胶电泳及凝胶扫描分析软件和数据统计,参照营养期、饥饿期、接合期皮层细胞骨架蛋白组分的组合和含量的变化,着重分析比较了接合后体发育期的皮层细胞骨架蛋白组分的变化,发现这一时期皮层细胞骨架蛋白组分中一部分蛋白含量持续降低,如225KD、150KD、120KD、88KD、84KD等;而另一部分蛋白组分的含量相对于接合期而言有所增加,如94KD、74-80KD、64KD、52KD、49KD等;并出现了205KD的特殊条带。这些变化,可能与接合后体发育期一方面延续了接合过程中皮层细胞骨架的特殊变化,另一方面向正常营养细胞循环恢复的过渡性特征有关。

Studies on Cortical Cytoskeletal Proteins at Exconjugants'Development Stage of Tetrahymena thermophila

Abstract:To study the changes of cortical cytoskeletal proteins at exconjugants' development stage of Tetrahymena thermophila , we used the cells of mating type- Tetrahymena thermophila BF 1 and BF 5 . When the conjugation course was finished ,the cells separated and became the exconjugants. Refeeding is required for the exconjugants undergo their first postzygotic cell cycle. We determinated the proper time for extracting cortical cytoskeletal proteins at exconjugants' development stage of Tetrahymena thermophila , according to the changes of cellular nucleuses with microscopic examination. Cortical cytoskeletal proteins were prepared by biochemical extraction with TritonX-100,SDS-PAGE and scan. The data statistics had been analyzed by a computer. The results showed that the content of some proteins continued reducing .for instance,225KD, 150KD, 120KD, 88KD, 84KD while the content of others became more than that of the content of conjugation stage e.g.94KD,74-80KD,64KD, 52.5KD,49KD. The 205KD only appeared in exconjugants' development stage. The changes of these proteins were involved in cortical reorganization during exconjugants' development stage . They also had some connection with the resuming of normal vegetative cell cycle.
