白芥子多糖的提取与含量测定

摘    要:用水提醇沉法提取白芥子多糖,用苯酚—硫酸比色法测定多糖含量.采用单因素和正交实验L9(33)考察了提取温度、提取时间、料液比、浸提次数和粉末粗细对白芥子多糖提取率的影响,从而确定出最佳的提取方案,并测定白芥子粗多糖的含量.结果表明,最佳提取工艺为A3B2C3,即在温度98℃下,料液比为1∶25,水提取2 h;此外,单因素实验得,浸提2次,粉末为40目,加5倍量95%乙醇醇析为宜;测得白芥子中多糖含量为3.821%,平均回收率为97.86%,相对标准偏差为RSD%=0.19,(n=5);各因素对白芥子多糖的提取率影响是不均等的,影响程度依次,为提取温度>料液比〉提取时间.
