植物生长调节剂对黄芪愈伤组织中次生代谢物质含量的影响

摘    要:以膜荚黄芪幼叶为外植体,运用正交设计研究2,4-D、6-BA和NAA等3种植物生长调节剂组合对愈伤组织生产黄芪黄酮和黄芪皂苷的影响.结果表明对黄芪黄酮合成的影响顺序是:NAA〉6-BA〉2,4-D;对黄芪皂苷合成的影响顺序是:2,4-D〉6-BA〉NAA.生产黄芪黄酮和黄芪皂苷的最佳培养基分别为;MS+2,4-D(0.5mg·L^-1)+6-BA(1.0mg·L^-1)+NAA(2.0mg·L^-1和MS+2,4-D(2.0mg·L^-1)+6-BA(2.0mg·L^-1)+NAA(1.0mg·L^-1).细胞伸长有利于愈伤组织中次生代谢物质的合成.

Abstract:Taking the leaves of Astragalus membranaceus ( Fisch. ) Bge. as explants, the effects of 2,4- D,6-BA and NAA on production of flavonoids of astragalus and saponins of astragalus were studied by using orthogonal design. The yield of total flavonoids of astragalus and total saponins of astragalus were used as criteria of estimating the efficiency of three regulators, the results showed that the effects of regulators on total flavonoids of astragalus content was as : NAA 〉 6-BA 〉 2,4-D, and the effect of regulators on total saponins of astragalus was as: 2,4-D 〉 6-BA 〉 NAA. The optimal culture medium for production of total flavonoids of astragalus was : MS + 2,4-D (0.5 mg·L^-1) + 6-BA ( 1.0 mg·L^-1 ) + NAA (2.0 mg·L^-1 ), and that for production of total saponins of astragalus was as : MS + 2,4-D (2.0 mg·L^-1 ) + 6-BA(2.0 mg·L^-1) + NAA( 1.0 mg·L^-1). Cells elongation facilitated the synthesis of secondary metabolites in callus.
