摘    要:本文应用二株抗角蛋白单克隆抗体(MAb D10为抗爪蟾角蛋白,MAb AF5为抗人角蛋白)对爪蟾卵母细胞、受精卵、不同发育期胚胎作免疫转移印迹观察,研究其角蛋白多肽亚单位的组成。发现不同发育期胚胎角蛋白表达有变化。同时应用免疫组织化学染色技术研究角蛋白在胚胎中的分布。发现角蛋白分布于表皮、脊索、嗅板、垂体原基、粘腺、咽上皮、神经管内表面等部位。

Abstract:Two anti-keratin MAbs were used to identify keratin expressed in embryos of Xenopus laevis. Mice anti-Xenopus keratin MAb D10 prepared in our Lab. Mice anti-human keratin MAb AF5 prepared in Chonqing Medical University. MAb D10 recognized5 polypeptides of keratin in fertilized eggs with Mr of 68, 62, 58, 56 & 52 Kd, while gastrula and neurula stages 58 Kd keratin disappeared. At tailbud stage-the period of formation of the primary organ rudiments, 5 additional types of keratins were expressed (64, 60, 49, 46 & 44 Kd), 40 stage was just the same. At 46 stage tadpoles, contained6 distinct polypeptide bands with molecular weights of 68, 64, 60, 52, 46 & 44 Kd keratins, while 62, 56 & 49 Kd disappeared.The adult epidermis of Xenopus contained 66, 60, 56 & 51 Kd keratins, in which 2 of them (60 & 56 Kd) appeared in the early embryos.This paper detected keratins of Xenopus embryo using mice anti-human keratin MAb AF5.The results were summarized in Table 1.Enzyme immunohistochemical Staining showed that keratins exist in Xenopus embr-yoic (St.28-46) notochord, epidermis, cement gland, pituitary, olfactory, pharynx and the inner surface of the neural tube.
