摘    要:建立了用离子色谱测定海带硫酸多糖硫酸根的方法。海带经酶解,季胺盐沉淀,离子交换和凝胶过滤,从中提取分离到3种多糖:F1,F2,F3,在对3个组分进行了GPC及红外光谱分析,证明匀为均一硫酸多糖的基础上,2组分分别经灰化,定容处理后,利用离子色谱对它们的硫酸根含量分别进行了测定,结果分别为:F1:28.41KD,F2:15.87KD,F3:32.50KD。与传统硫酸钡浊度法相比,该方法消除了其它离子及杂质的干扰,操作简单,具有精确,灵敏,高效等许多优点,是测定硫酸多糖硫酸根含量的理想方法。

Abstract:An ion-chromatographic method for the determination of sulfate group of sulfated posaccharidesfrom laminaria was described . Three polysaccharide fractions (F1 , F2 , F3) were separated from laminaria with enzymatic lysis precipitation by quaternary ammonium , ion exchange and gel filtration . After three fractions were shown to be homogenous sulfated polysaccharides by GPC and IR , the determination of sulfate group was performed following combustion by ion-chromatography respectively . The sulfated group content of F1 , F2 , F3 was 28.41% , 15.87% , 32.50% , respectively. Due to removing interference from other ions and impurities , the ion-chromatographic method was accurate , sensitive , quick and simple comparing with turbidimetry , so this is an ideal method for the determination of sulfate group of sulfated posaccharides .
