阿霉素对人胃癌细胞SGC-7901增殖和凋亡的影响

观察阿霉素对SGC-7901细胞的凋亡诱导作用.采用台盼蓝拒染法测定细胞生长抑制率,计算半数抑制质量浓度值;采用DNA琼脂糖凝胶电泳技术检测细胞凋亡,倒置显微镜观察细胞形态结构的变化;采用流式细胞仪检测阿霉素对细胞周期的影响,探讨阿霉素对SGC-7901细胞增殖和凋亡的影响.结果表明,阿霉素对SGC-7901细胞具有抑制作用,IC50为5.7μg.mL-1;DNA琼脂糖凝胶电泳有典型的梯形条带,倒置显微镜观察显示,阿霉素能抑制细胞增殖,可致细胞形态学改变,出现明显的凋亡形态学特征,流式细胞仪检测出现凋亡峰.说明阿霉素能明显抑制人胃癌细胞株SGC-7901细胞增殖,并能诱导其凋亡.

Effect of ADM on proliferation and apoptosis of human gastric cancer cell SGC-7901

To observe the effect of ADM on cells apoptosis of SGC-7901 cells.The SGC-7901 cells were treated by ADM and the inhibitory ratio of cells was measured by trypan blue stain assay,the IC50 value was calculated.Cells apoptosis were detected by DNA agarose gelelectrophoresis.The cell cycles were analyzed by flow cytometry system after treatment with ADM.Morphologic changes were observed using phase-contrast microscopy.The results show that the SGC-7901 cells proliferation were remarkably inhibited by ADM.The IC50 values were 5.7 μg·mL-1.The typical DNA ladder on agarose gel electrophoresis for analysis of cellular apoptosis were significantly appeared.ADM could restrain the SGC-7901 cells proliferation,and to cause the morphologic changes of apoptosis.Apoptosis peaks appeared with flow cytometry analysis.ADM can significantly inhibit of human gastric cancer SGC-7901 cells proliferation,induce cell apoptosis.
