| 摘 要： | 核糖核酸的制备和分析受多种因素的影响．因此，在特定条件下制备高纯度和完整性好的ＲＮＡ的方法研究至关重要．本文以大白鼠肝为材料，研究ＲＮＡ的制备方法，建立了有效的ＲＮＡ制备方法．用紫外分光光度计和变性电泳分析所制备的ＲＮＡ，发现其纯度Ａ２６０Ａ２８０＝１．７～２．０，２８ＳｒＲＮＡ与１８ＳｒＲＮＡ的带宽比为２１．本方法具有节约节时，重复性好，产量较高，无ＤＮＡ污染等优点．这为ｃＤＮＡ合成，基因表达和调控等研究奠定了基础．

| Abstract: | The preparation of RNA is effected by various factors. So it's very important to study an efficient method for preparation of complete RNA with high purity. In the paper,white rat's liver was used as material in order that the method for preparing RNA was established. The purity of prepared RNA amounted from 1.7 to 2.0by the analysis of the UV spectrophotometer. The width of 28SrRNA lane∶the one of 18SrRNA lane was 2∶1,tested by the denatured agrose gel electrophoresis. The method had several advantages such as easy repeat,economy,high yield of RNA,DNA-free contamination,etc.This laid the foundation of cDNA synthesis, gene regulation control and so on. |
