同型半胱氨酸对血管抗氧化酶基因表达的影响

摘    要:目的探讨同型半胱氨酸(Hcy)诱导抗氧化酶基因表达的影响。方法采用实时定量RT-PCR测定基因的表达,用DCF—DA荧光探针测定细胞内ROS;研究材料为HUEVC及高Hcy小鼠胸主动脉;结果同型半胱氨酸能诱导人脐静脉血管内皮细胞(HUEVC)的ROS水平升高,抑制HUEVC的谷胱甘肽过氧化物酶(GSHPx)和过氧化氢酶(CAT)基因的表达。在高同型半胱氨酸血症小鼠胸主动脉中,谷胱甘肽过氧化物酶、过氧化氢酶和锰超氧化物歧化酶(Mn-SOD)基因表达均明显降低。结论 Hcy能促进上皮细胞内活性氧的产生,可明显促进氧化损伤,同时显著降低抗氧化酶(谷胱甘肽过氧化物酶、过氧化氢酶及锰超氧化物歧化酶)的活性。
