黄瓜离体雌核发育早期的特异蛋白研究

摘    要:通过改进的IEF-SDS-PAGE技术研究了黄瓜雌核发育早期的特异蛋白质表达,并发现了11个特异多肽点,其中3个(A 1~A 3)可能是开启分裂基因表达的一类反式作用因子,起到抑制该基因表达作用.8个(B 1~B 8)可能是启动雌配子分裂密切相关的基因表达产物,以上特异蛋白在多次重复中均显示出良好的重复性,因此认为,这些特异蛋白可能对黄瓜雌核发育早期的启动起重要作用.

Abstract:Expression of specific proteins during early stage of cucumber in vitro gynogenesis was investigated using modified IEF-SDS-PAGE technique.11 specific protein dots were found.3 protein dots including A1(21.0 kDa,PI6.5),A2(20.0 kDa,PI6.6),A3(11.5 kDa,PI7.5) and were found to be specific in the ovary before culture,which are probably the factors which inhibit the genes from initiating gamet cell division.8 protein dots including B1(37.6 kDa,PI4.5),B2(37.6 kDa,PI4.6),B3(24.3 kDa,PI6.3),B4(19.9 kDa,PI6.3),B5(27.5 kDa,PI6.7),B6(18.3 kDa,PI6.9),B7(16.2 kDa,PI7.0),B8(37.7 kDa,PI8.2) were found to be specific after 3 days of culture,which are probably the products of related genes that initiate gamet cell division.These specific proteins showed good repeatability in the repeat experiments.It is therefore suggested that these specific proteins play an important role in cucumber in vitro gynogenesis.
