薏苡的组织培养及其过氧化物酶分析

摘    要:薏苡胚轴外植体在附加2.4—D4mg/L(单位下同)的 N_6培养基中形成愈伤组织;转至2.4—D0.5中,愈伤组织迅速增殖,出现根、芽分化;在2.4—D4中继代培养的愈伤组织则逐渐老化,并丧失分化能力;外植集在 BA2的培养基中愈伤组织增殖缓慢、褐化、无分化能力。比较了此四种愈伤组织的过氧化物酶活性及其同工酶谱,前二者的过氧化物酶活性明显高于后二者,且同工酶谱多两条酶带—E_6、E_7。

Abstract:Calluses were produced from hypocotyl explant of Coix la- cryma-jobi cultured on N6 medium with 2.4-D4mg/L.when trans- sferred to the medium with 2.4-D 0.5mg/L,the calluses grew rapidly and differentiated in to buds and roots,but if subcul- tured on the medium with 2.4-D4mg/L,they would be aged grad- ually and lost differentiation potential.The calluses derived from hypocotyl explant cultured on medium with BA2mg/1 grew slowly and became browning and no bud and root regene- ration occurred.Comparing the four calluses,the first two show- wed higher peroxidases activity than the others and there were two extra peroxidase isozyme bands E_6 and E_7,typical of the calluses with differentiation capacity.
