人类逆转录病毒的长末段重复序列(VRTVLTR)的表达与肺癌转移的关系

摘    要:肺癌是最常见的恶性肿瘤之一。肿瘤癌很容易发生转移。实验以17例肺癌(5例肺腺癌、1例腺鳞癌、7例肺鳞癌及4例小细胞末分化癌)的原发灶、癌旁正常组织及转移淋巴结为材料,采用PCR及非放射性标记的RNA斑点杂交分析LTR在基因组的存在及其表达情况。研究结果发现:LTR序列在17例肺癌病人的正常组织及肿瘤组织的基因组中普遍存在,LTR致肺癌转移与其插入基因组中无关;LTR致肺癌转移与其表达增高有关,而且其高表达与小细胞肺癌的转移无关,而与非小细胞肺癌的转移密切相关。提示LTR参与了非小细胞肺癌的转移过程。实验结果为从细胞系获得的结论提供了进一步的证据。

Abstract:Lung cancer is one of the most common malignant tumors in human, and lung adenoc arcinoma is easier to metastasize than the other kinds of lung cancers. Two lung adenocarcinoma cell lines, AGZY S3a and Anip973, which are originat ed from the same cell line but manifested different metastasis potential, are su bjected to mRNA differential display in our laboratory to analyze the difference s in the gene expression between the two cell lines. Significant differences be tween the two cell lines were obtained. One of the differentially expressed frag ment , the long terminal repeats of human retrovirus gene (HRTV LTR) is highly e xpressed in Anip973 while low expressed or even not expressed in AGZY 83a. The result suggested that the expression of LTR may involve in the process of meta stasis of lung adenocarcinoma. In the present study, primary tumors, normal tiss ues and metastatic lymph nodes of seventeen lung cancer patients were obtained t o analyze the expression of LTR with the technique of polymerase chain reaction and RNA Dot Hybridization with non radiative labeled probe. The result showed t h at LTR universally existed in the genome of normal tissues and tumor tissues of seventeen patients and its integration has no relation with the metastasis of l ung cancer. However, the over expression of LTR was responsible for the metasta sis of non small cell lung cancer but not for small cell lung cancer, which ind icated that LTR may involve in the process of metastasis of non small cell Lun g cancer.
