宁夏枸杞愈伤组织器官发生和体细胞胚发生过程中过氧化物酶和酸性磷酸酶同工酶的研究

摘    要:利用已建立的宁夏枸杞(Lycium barbarum L.)叶愈伤组织器官发生和体细胞胚发生体系,比较研究了两条离体再生途径早期阶段愈伤组织的生长特性、过氧化物酶(POD)和酸性磷酸酶同工酶的活性及酶谱的变化。结果表明:(1)与继代愈伤组织的“S”形生长曲线不同,器官发生和体细胞胚发生过程的愈伤组织生长延迟期不明显,且第3d后一直处于指数生长期,培养后期器官发生的愈伤组织生长速率快于体细胞胚发生;

Abstract:By the established sys te m of organogenesis and somatic embryogenesis of Lycium barbarum (L.) calli, a comparative study on the changes of callus growth, activities and isoenzyme pa tterns of peroxidase (POD) and acid phosphatase in the early stages of the two i n vitro regeneration pathways was carried out. The results were as follows: (1) Unlike the "S" shaped growth curve of subcultured calli, the stationary phase o f calli during organogenesis and somatic embryogenesis was indistinct and after 3 days of culture, calli were always in the exponential phase of growth. In the later stages of callus differentiation, the growth rate of callus for organogene sis was faster than that for somatic embryogenesis. (2) POD activities in these two regenerative pathways shared similar changes and had two peaks, respectively . (3) Both P 3 and P 4 bands of POD isozyme were correlated with organogenesis and somatic embryogenesis. P 6 band marked the development of embryogenic cell s while P 5 band was expressed during the formation of both embryogenic cell a nd meristemoid. (4) A 3 band of acid phosphatase might be viewed as the marker of adventitious bud and globular embryo formation during the late morphogenetic period.These results showed that the isoenzyme profiles of POD and acid phosphat ase can be proposed as a rapid and sensitive biochemical marker system of differ entiation trends (organogenesis and somatic embryogenesis) during in vitro m orphogenesis of L. barbarum.
