甘薯毛状根植株再生研究

摘    要:用发根农杆菌R1000菌株感梁甘薯(渝薯34品种),获得转化毛状根,从毛状根中选择出了5个无性系,当毛状根培养在含有2mg/L6-BA+1mol/L玉米素的MS培养基上诱导出了愈伤组织,当愈伤组织培养在含有1mg/LKT+1mg/L6-BA+0.5mg/LIAA的培养基上,3周以后获得了再生植株,用随机及增多态性DNA(RAPD)分子标记检测出了再生植株的多态笥。

Abstract:Transformed hairy roots were induced in sweet potato ( Ipomoea batatas ) by Agrobacterium rhizogenes strain R1000.Five clones were selected from hairy roots.Callus was induced when hairy roots were cultured on the medium containing 2 mg/L 6 BA and l mg/L zeatin.Plantlets were regenerated from callus in 3 weeks on the media containing l mg/L KT,l mg/L 6 BA and 0 5mg/L IAA.The polymorphism of regenerated plantlets was detected by the use of a RAPD marker.
