小鼠短途运输应激及其营养干预的研究

摘    要:目的研究实验小鼠的短途运输应激反应并研制用于缓解该应激的营养干预剂。方法实验1采用BALB/c小鼠和ICR小鼠,经1.5 h运输后返回原处,分析运输后即刻(0 h)、24 h、48 h、72 h的血糖(GLU)、外周白细胞总数(WBC)、血清中皮质酮(CORT)、白细胞介素2(IL2)和β-内啡肽(β-EP)的变化,评价短途运输应激对2种小鼠的影响。实验2采用BALB/c小鼠以运输全程放置营养补充剂的方式进行运输干预实验,测定并比较干预组和运输对照组及空白对照组的应激相关指标,由此评价干预效果。结果在实验1中,运输后即刻2种小鼠GLU、CORT、IL2、β-EP、WBC均降低,但于运输结束后72 h内陆续恢复;实验2中,营养干预组GLU、β-EP、IL2和WBC的恢复时间均比运输对照组提前约24 h。结论短途运输可导致小鼠体能过度消耗以及神经-内分泌-免疫紊乱,在常规饲养条件下进行自然恢复至少需要72 h,运用营养补充剂进行运输同步干预有助缩短小鼠运输后恢复所需时间。

Abstract:Objective To investigate the short-term transportation stress of laboratory mice and develop nutritional intervention that can be used to relieve the stress.Methods In the first experiment,BALB/c mice and ICR mice were transported by road for 1.5h,short-term transportation stress in the two different strains were accessed by the alteration of GLU,WBC,serum level of CORT,IL-2 and β-EP in the time of 0 h,24 h,48 h,72 h after transportation.In the second experiment,BALB/c mice were transported in the container equipped with nutrition supplementation,and then the stress related indicators were measured and compared across intervention group,transporting group and control group to evaluate the intervention effect.Results In the first experiment,both strains showed decreased GLU,CORT,IL-2,β-EP and WBC at the termination of transportation,but all the indicators return to normal within 72 h.In the second experiment,GLU,IL-2,β-EP and WBC in invention group resumed almost 24 h earlier than transporting group.Conclusions Short-term transportation would be exhausting and trigger neural-endocrine-immune disorder in mice;it needs at least 72 h for mice to recover in the routine condition,while giving nutrition supplementation at the termination of transportation would contribute to a shorter recovering period
