葎草抗炎镇痛有效部位筛选及机制

摘    要:目的 筛选葎草镇痛抗炎的有效部位,初步探讨有效部位镇痛抗炎的作用机制。方法 采用溶剂萃取法将葎草水提液分离为不同部位;采用小鼠热板和醋酸扭体疼痛模型,筛选葎草镇痛作用的有效部位;采用二甲苯小鼠耳肿胀、大鼠蛋清足跖肿胀2种急性炎症模型以及大鼠棉球肉芽肿慢性炎症模型,筛选葎草抗炎作用的有效部位;检测有效部位对醋酸所致疼痛模型小鼠血清前列腺素E2(PGE2)、环磷酸腺苷(cAMP)含量以及蛋清所致大鼠足跖炎症组织NO和PGE2含量的影响。结果 葎草石油醚萃取部位明显延长热板所致小鼠的痛阈值(P < 0.05),明显减少醋酸所致小鼠扭体反应次数(P < 0.05);葎草石油醚萃取部位明显抑制二甲苯所致小鼠耳廓肿胀(P < 0.05),明显抑制蛋清所致大鼠足跖肿胀和大鼠棉球肉芽肿(P < 0.05)。葎草石油醚萃取部位明显降低醋酸所致疼痛小鼠血清PGE2、cAMP的含量(P < 0.05),明显降低蛋清所致大鼠足跖组织NO和PGE2含量(P < 0.05)。结论 葎草的水提液的石油醚萃取部位是镇痛抗炎的有效部位,其镇痛作用机制可能与降低PGE2、cAMP的含量有关,其抗炎作用机制可能与减少炎症组织NO和PGE2含量有关。

Abstract:Objective:To screen the analgesic and anti-inflammatory effective fractions from Humulus scandens, and to study the relative mechanism. Methods: Water extract of H. scandens were respectively extracted into different fractions by using solvent extraction method. The analgesic effects of the effective fractions from H. scandens were observed by hot plate and acetic acid writhing model. Meanwhile, the acute inflammatory models, such as xylene-induced ear swelling in mice, egg white-induced paw swelling in rats, and the chronic inflammatory model granuloma induced by cotton pellet implantation in rats, were used in researching the anti-inflammatory effects of the effective fractions. To explore the analgesic and anti-inflammatory mechanism, the serum Prostaglandin E2 (PGE2) and Cyclic Adenosine monophosphate (cAMP) levels in mice with pain induced by acetic acid were determined, and the inflammation caused by egg white foot plantar organization NO and PGE2 levels in rats were also determined. Results:Treated with with petroleum ether fraction, the pain threshold with hot plate method was significantly prolonged (P < 0.05), the writhing number was significantly reduced (P < 0.05), the ear and paw swelling was significantly inhibited (P < 0.05) and the granuloma was significantly decreased in rats (P < 0.05). The petroleum ether fraction decreased significantly the serum PGE2 and cAMP levels in mice with pain induced by acetic acid (P < 0.05), and decreased significantly the inflammatory caused by egg white foot plantar organization NO and PGE2 levels of rats (P<0.05). Conclusion: The petroleum ether fraction of H. scandens is analgesic anti-inflammatory effective fractions, and its mechanism of analgesic effect may be related to the reduction of PGE2 and cAMP levels, and its mechanism of anti-inflammatory effect may be related to the reduction of NO and PGE2 levels in the inflammatory tissue.
