摘    要:对苦菜多酚提取工艺进行优化研究.用单因素试验对提取溶剂进行筛选,结果表明乙醇和一定浓度的盐酸组成的复合体系是理想的提取溶剂,用析因试验、最速上升试验、中心组合实验对提取条件进一步优化.结果发现乙醇浓度、提取温度、提取溶剂的pH、料液比都对苦菜多酚提取产生一定的影响,其中pH和乙醇浓度的影响最为显著.最佳的多酚类化合物提取条件为料液比1∶30,提取溶剂为65%乙醇,其酸度为0.78 mol/L,提取温度60℃,此时苦菜多酚的得率为19.12 mg/g.

Abstract:Extraction of polyphenols from Sonchus branchyotus DC.was optimized.The composed system that contained ethanol and a certain concentration hydrochloric acid was the ideal extracting solvent by single factors experiment.The extracting condition was further optimized with factorial,the steepest ascent and central composite design.The results showed that the extracting rate of polyphenols was affected by the ethanol concentration,ration between material and solvent,extracting time and temperature,of which pH and ethanol concentration had the most significant effect on the extracting rate.The optimal solvent,temperature,acidity of solvent,ratio between material and solvent was 65% ethanol,60℃,0.78 mol/L and 1∶30,respectively.Under the afore-mentioned conditions the extraction rate of polyphenols was 19.12 mg/g.
