电泳羟基磷灰石色谱吸附BSA机理

摘    要:将电泳与羟基磷灰石色谱技术耦合 ,开发了一种新的电泳色谱技术。建立了羟基磷灰石 (HAP)吸附牛血清白蛋白 (BSA)的物理模型和数学模型 ,建立了不同的实验装置以研究电场下 HAP吸附 BSA的反应动力学特性以及电动色谱吸附 BSA过程的传质动力学特性。实验结果表明 ,施加电场不会改变吸附反应的平衡常数与速率 ,但在电泳羟基磷灰石色谱装置中 ,施加电场在羟基磷灰石颗粒表面产生的电渗流能够加强扰动 ,减少液膜扩散阻力 ,提高羟基磷灰石色谱的动态吸附容量

Abstract:A novel kind of preparative electrophoretic chromato graphy, electrophoretic hydroxylapatite chromatography (EHAC), was developed by coupling electrophoresis with hydroxylapatite chromatography. A physical and mathematical model was developed for the adsorption of Bovine Serum Albumin (BSA) on Hydroxylapatite (HAP) with two experimental installations to study the mass transfer kinetics of the process in an electric field. The experimental results show that the equilibrium and adsorption rate are not altered by the electric field. However, the electroosmotic flux on the bead surface more effectively disturbed the liquid flow, which reduced the mass transfer resistance and, consequently, increased the available bed volume for adsorption and the dynamic binding capacity in separation by EHAC. The study provides the theoretical basis for further application of EHAC.
