乳腺癌组织芯片的应用与HER-2/neu的表达

目的 :构建乳腺癌及乳腺癌 -良性病变组织微阵列 ,研究HER 2 /neu在原发性乳腺癌和乳腺良性病变中的表达并探讨HER 2 /neu的判断标准。方法 :制作乳腺癌 -乳腺良性病变组织微阵列 (tissuemicroarray)。用免疫组织化学方法检测HER 2 /neu在 180例乳腺癌 ,32例乳腺良性病变组织中的表达。结果 :构建乳腺癌及乳腺癌 -乳腺良性病变组织微阵列 4个 ,180例乳腺癌中HER 2 /neu阳性率为 37.78% (6 8/ 180 ) ,32例乳腺良性病变不表达。乳腺癌组织中HER 2 /neu的过表达率显著高于乳腺良性病变组织 (P <0 .0 5 )。结论 :乳腺癌 -乳腺良性病变组织微阵列的建立使乳腺癌相关基因及其蛋白产物的筛选工作简便、快捷。HER 2 /neu的过表达与乳腺癌的发生密切相关

Application of Tissue Microarry of Breast Carcinoma and Expression of HER-2/neu

Objective:To develop tissue microarray of breast benign lesion and carcinoma, and investigate the role of HER-2/neu in the breast benign lesion and carcinoma and the standard of HER-2/neu scoring system.Methods: Tissue microarray of breast benign lesion and carcinoma was constructed by using a microarray. The expression of HER-2/neu was detected on the section of tissue microarray by immunohistochemistry.Results: Breast benign lesion and carcinoma tissue microarray was developed. The positive rate of the expression of HER-2/neu in 180 cases with breast cancer was 37.78%(68/180). None of the 32 cases with breast benign lesion express HER-2/neu protein. There were significant differences of overexpression of HER-2/neu between breast benign lesion and breast carcinoma(P< 0.05). Conclusions:The development of tissue microarray of breast benign lesion and carcinoma facilitates surveys of breast cancer-related gene candidates. The overexpression of HER-2/neu is closely involved in the carcinogenesis of breast carcinoma.
