拟南芥FIE基因特异片段的克隆及序列测定

摘    要:根据拟南芥FIE基因的序列设计PCR引物,以拟南芥基因组DNA为模板,通过PCR反应扩增FIE基因特异性片段,扩增产物经琼脂糖凝胶电泳分析,长为1633bp,与预期结果相符。将该基因片段定向克隆到pGEM-T载体上,得到重组质粒p1633,酶切鉴定及测序分析结果表明该克隆为拟南芥FIE基因特异片段,与拟南芥FIE基因比较同源性达99.8%,可以作为探针使用。

Abstract:According to the sequence of Arabidopsis thaliana FIE gene, PCR primers were designed. FIE specific fragment was amplified on the model of Arabidopsis thaliana DNA. The product was 1633 bp in length by gelose gel electrophoresis and same to the prospective result. Linked to pGEM-T Vector, the recombinant plasmid was p1633. Restriction analysis and sequence analysis had proved that the clone product was the specific fragment of Arabidopsis thaliana FIE gene. Compared with Arabidopsis thaliana FIE gene, the cloned FIE fragment shared 99.8% homology at nucleotide level. The specific fragment can be used as probes.
