凝固酶测定试验鉴定葡萄球菌的影响因素

目的 探讨玻片法和试管法检测大酶的影响因素方法。方法 按标准方法操作比较两种血浆（人血浆和兔血浆）及三种抗凝剂（ＥＤＴＡ－Ｎａ２，ＥＤＴＡ－Ｋ２，柠檬酸钠）对结果的影响。结果 使用柠檬酸钠和ＥＤＴＡ－Ｎａ２抗凝的兔血浆和人血浆，８株金黄色葡萄球菌均产生葡萄球菌凝固酶和聚集因子，而其它２６株葡萄球菌包括表皮葡萄球菌、溶血葡萄球菌、腐生葡萄球菌和人葡萄球菌不产生葡萄球菌凝固酶和聚集因子。结论 柠檬酸钠

Coagulase Test for Identification of Affecting Factors of Staphylococci

Objective To explore coagulase test for the identification of affecting factors of staphylococci through the slide test and the tube test.Methods On the standard procedure,to study the effects of two different plasma including human plasma and rabbit plasma and the effects of three different anticoagulants including EDTA.Na 2,EDTA.K 2 and sodium citrate on the results of the test.Results All 8 strains of S.aureus produced staphylocoagulase and clumping factor using rabbit plasma and human plasma containing sodium citrate and EDTA.Na 2,the other 26 strains of staphylococcus spp including S.epidermidis, S.haemolyticus, S.saprophyticus and S.hominis did not produced them.Conclusion Correct results could be obtained using either human plasma or rabbit plasma containing EDTA.Na 2 and sodium citrate in determining staphylococcal coagulase,but human plasma containing EDTA.K 2 anticoagulant could not be used to detect free coagulase (staphylocoagulase).
