甘肃张掖地区葡萄天然酵母菌的筛选及其系统发育分析

摘    要:目的:通过对张掖地区酵母菌的筛选,了解张掖地区酵母菌的生物多样性。方法:采用YPD培养基分离纯化菌株,通过26SrDNA基因D1/D2区序列分析确定菌种的系统进化地位。结果:分离筛选出6个属天然酵母菌。分别为:假丝酵母属Candida、Meyerozyma属、红酵母属Rhodotorula、孢汉逊属Hanseniaspora、酿酒酵母Saccharomycescerevisiae和毕赤酵母属Pichia,共10个种,其中包括6个疑似种。

Abstract:Objective:The diversity of culturable yeast strains in GanSu Province of China was investigate. Not only isolation and molecular phylogenetic analysis were performed in order to expand our knowledge on diversity of yeasts, but also efforts focused on screening for natural yeasts. Methods:Yeasts were detected on YPD, respectively. Identity and genetic diversity of strains isolated was determined by spatial 26S rDNA gene D1/ D2 sequences and rep-PCR fingerprint. The physiological tests were carried out to determine the phonotypic differences between strains showing high similarity of 26S rDNA gene D1/D2 sequences.Results:Separation screen out six belongs to natural yeast, respectively: Candida species Candida, Meyerozyma genera and Rhodotorula Rhodotomla, spore han amazonian belong to Hanseniaspora, Saccharomyces cerevisiae Saccharomyces cerevisiae and Pichia Pichia, a total of 10 species, including six of suspected.
