根癌农杆菌电击转化条件的研究

摘    要:探索了根癌农杆菌EHA105的高效转化方法.将双元载体质粒pBIN19/glgB经电击转化法导入根癌农杆菌EHA105,研究不同实验条件对转化率的影响.结果表明:在细胞浓度OD600为1.0时进行电转化,转化率最高,达到每微克DNA1.12×105CFU;当电场强度为11 kv/cm,转化率最高,达到每微克DNA1.78×105CFU;在一定的细胞浓度范围内,电击转化率与细胞浓度密切相关,转化率随着细胞浓度的上升而上升;质粒大小能影响电击转化率,转化率随着质粒的增大而下降.应用该优化的电击转化条件将双元载体质粒pBIN19/glgB成功导入根癌农杆菌EHA105,构建了植物双元表达载体.

Abstract:To explore effective transformation methods for Agrobacterium tumefaciens EHA105.The binary vectors pBIN19/glgB was introduced into Agrobacterium tumefaciens EHA105 by electroporation.Effects of different electroporation conditions on transformation frequency were studied.Transformation frequency reached to1.12×105 CFU/μg DNA,the highest level,when OD600 was at about 1.0.Transformation frequency also reached a highest level of 1.78×105 CFU/μg DNA when the electrical field strength was 11 kv/cm.The transformation frequency was related to cell density,it increased with the increase of cell concentration.The size of plasmid can effect transformation frequency,it decreased with the increase of the size of plasmid.The binary vectors pBIN19/glgB was transformed into Agrobacterium tumefaciens EHA105 by this modified protocol.
