摘    要: 为研究冠突散囊菌与金黄色葡萄球菌混合发酵次级代谢产物及细胞毒活性,采用硅胶柱、凝胶柱和高效液相色谱等方法进行分离纯化,并通过各种波谱技术进行结构鉴定,以4种肿瘤细胞A549、MCF-7、Bel-7402、Colo205为供试细胞株,采用MTT法对化合物进行细胞毒活性研究。从冠突散囊菌与金黄色葡萄球菌固体混合发酵提取物中分离鉴定了7个化合物,其中4个吲哚二酮哌嗪生物碱为echinulin(1)、cristatumin F(2)、neoechinulin A(3)、preechinulin(4),2个聚酮类化合物为flvoglaucin(5)、tetrahydroauroglaucine(6),1个nonadride类化合物为epiheveadride(7)。活性结果表明,化合物对4种肿瘤细胞株均具有不同程度的细胞毒活性,化合物3、5和7对肝癌Bel-7402细胞毒活性较强。

Abstract:To evaluate the cytotoxic activity of secondary metabolites from co-culture of Eurotium cristatum with Staphylococcus aureus, seven known compounds, including echinulin(1), cristatumin F(2), neoechinulin A(3), preechinulin (4), flvoglaucin (5), tetrahydroauroglaucine (6), and epiheveadride (7) are isolated from the solid culture of the co-culture of Eurotiumcristatum with Staphylococcus aureus. The structures are checked on the basis of spectroscopic analysis NMR experiments and by mass spectrometric measurements. These metabolites are evaluated in vitro for cytotoxicity activities against four tumor cell lines such as A549, MCF-7, Bel-7402, and Colo205. Compounds 3, 5 and 7 are found to show moderate cytotoxicity against the Bel-7402 cancer cell line.
