祛痰止咳颗粒指纹图谱研究

摘    要: 采用HPLC法构建了祛痰止咳颗粒指纹图谱,色谱条件为Elite Hypersil BDS(4.6 mm×250 mm,5 μm)色谱柱,以甲醇(A)-0.15%(〖WTBX〗φ〖WTBZ〗)冰醋酸溶液(B)为流动相,线性梯度洗脱A(15%→75%),流速1.0 mL/min,柱温25 ℃,检测波长260 nm。该指纹图谱中,确定了19个共有峰,并采用对照品对照法及HPLC-ESI-MS/MS联用技术鉴定出10个黄酮类化合物。构建的指纹图谱,操作简便、准确可靠、重复性较好,为祛痰止咳颗粒质量控制提供了科学依据。

Abstract:HPLC fingerprint of Qutanzhike Granule was established with Elite Hypersil BDS column(4.6 mm×250 mm,5 μm),methanol(A) and 0.15% acetic acid(B) as mobile phase.The elution profile was: 0~95 min 15% to 75% A in B(linear gradient). The column temperature was set at 25 ℃ and the flow rate was 1.0 mL/min. The detective wavelength was at 260 nm. 19 common peaks were selected as the fingerprint peaks from 12 batches of the production. In the established fingerprint, 10 flavonoids were identified by the HPLC-ESI/MS/MS based on MS spetra and standard reference. The method can be used for quality control of Qutanzhike Granule with great precision,accuracy and good reproducibility.
