D-半乳糖致衰老小鼠模型的实验研究

摘    要:目的探讨应用D-半乳糖制作小鼠衰老模型,并进行指标检测.方法 D-半乳糖连续皮下注射42 d制作小鼠衰老模型,同时进行体质量变化测量、行为学测试和形态学衰老指标的测定.结果与正常对照组相比,造模4周后,D-半乳糖致衰老小鼠体质量显著降低(P0.01);D-半乳糖致衰老小鼠学习记忆能力减弱,水迷宫探索路径长度、搜台潜伏期均显著延长(P0.01);形态学观察显示各器官组织结构出现衰老改变.结论 D-半乳糖可以成功制作小鼠衰老模型.

Abstract:Objective To explore the application of D-galactose in establishing aging mice model,and to detect the related indexes. Method D-galactose was given to the mice continuously by subcutaneous injection for 42 days to make aging model. During this period,their changes in weight,behavior and senescence indications were measured as well. Results Compared to the control group, the body weight of the aging mice decreased significantly after 4 weeks modeling (P<0. 01). Besides,both exploration path length and escape latency were seriously prolonged in the water maze experiment (P<0. 01),indicating that the ability of learning and memory retarded in the aging mice. Morphological observations showed aging changes in the structures of several organs and tissues. Conclusion D-galactose can successfully establish aging mice model.
