| 摘 要： | 为了延长胰高血糖素样肽-1(GLP-1)在血浆中的半衰期,实验构建了编码GLP-1和白蛋白的融合蛋白基因,并在毕赤酵母中获得高效表达.发酵液经超滤后,分别采用了阴阳离子交换法和染料亲和法进行纯化.结果表明,用染料亲合法纯化能够有效地去除发酵过程中产生的弱酸性绿色化合物.经电泳鉴定纯度可达到95%,回收率可达到60%.

| Abstract: | To reduce the serum clearance of Glp-1,a chimeric gene encoding and an Glp-1/albumin fusion protein were expressed in Pichia pastoris effectively.After ultrafiltration,the ferment liquid was purified by ion-exchange chromatography and blue sepharose affinity chromatography.The result shows that the purification by blue sepharose affinity can remove the weak acidic green compound during the fermentation process effectively.The purity of the fermentation product could reach 95% by means of electrophoresis,and the total recovery yield could reach up to 60%. |
