黄荆子乙酸乙酯提取物诱导结肠癌HT-29细胞凋亡

目的研究黄荆子乙酸乙酯提取物（EVn-50）诱导人结肠癌HT-29细胞凋亡作用。方法用不同浓度的EVn-50处理体外培养的HT-29细胞；碘化丙啶（PI）染色流式细胞术（FCM）检测细胞凋亡率；琼脂糖凝胶电泳观察DNA梯形条带；Western blot分析Bcl-2和Bax蛋白表达。结果EVn-50以浓度依赖的方式增加HT-29细胞凋亡率（P〈0．05）。10．0tanoVLEVn-50处理HT-29细胞48h导致典型DNA梯形条带形成。10．0umol/LEVn-50以时间依赖方式下调Bcl-2蛋白表达，同时，上调Bax蛋白表达（P〈0．05）。结论EVn-50诱导HT-29细胞凋亡作用与增加Bax／Bcl-2比值有关。

Induction of apoptosis in human colon cancer HT - 29 cells by the acetoacetate extract of the vitex negundo seeds

Objective To investigate the effects of the acetoacetate extract of the Vitex negundo seeds（EVn - 50） on apoptosis of human colon cancer HT- 29 cells. Methods Human colon cancer HT - 29 cells cultured in vitro were treated with various concentration of EVn-50. The apoptotic effect was detected using flow cytometry （FCM） after PI staining. DNA ladder bands were observed by DNA agarose gel electrophoresis. The expression of Bcl - 2 and Bax protein were analyzed by Western blot. Results EVn - 50 significantly induced increase of the percentage of apoptotic cells（ P 〈 0.05）, in a concentration- dependent manner. The observation by DNA agarose gel electrophoresis found that the treatment with 10.0 gg/mL EVn - 50 for 48 h could produce DNA frag- mentation. EVn - 50（ 10.0 g/mL） a time - dependent reduced the expression of the Bcl - 2 protein and increased expression of Bax protein （ P 〈 0.05）. Conclusion EVn - 50 efficiently induces apoptosis of human colon cancer HT- 29 cells, and its mechanisms are involved in increase of the ratio for Bax to Bcl- 2 protein.
